PREPARATION AND STABILITY OF AMPICILLIN TRIHYDRATE AND EUDRAGIT-RS COPRECIPITATES FOLLOWING DISPERSION TECHNIQUE

Abstract

 ABSTRACT Ampicillin trihydrate is a broad spectrum antibacterial agent having short biological half-life and wide therapeutic index. These necessitated its formulation into sustained release dosage form by decreasing the dissolution rate using acrylic resin (Eudragit-RS(R)). The aim of this study is to prepare and asses the stability of ampicillin and Eudragit-RS(R) coprecipitates following dispersion technique. Coprecipitates of ampicillin trihydrate and Eudragit-RS(R) acrylic resin in the ratios of 2:1 (III) and 1:1 (II) were prepared by co precipitate technique. The stability studies of the coprecipitates of ampicillin tryhydrate and Eudragit-RS(R) were carried out by using thin-layer chromatography (TLC) and infra-red (IR) spectrophotometric techniques. The precipitated drug and the dispersed mixtures produced only one spot each on the TLC plate similar to that of the pure drug while, the carrier did not show any spot on the TLC plate. The resolution factor (RF) values for all the spots were about the same as that of the pure drug. The dissolution rate was slower for 1:1 than for 2:1 drug/polymer dispersions. The spectra of the precipitated drug and drug coprecipitates are similar to that of the untreated pure drug and no new peaks were recorded indicating absence of any possible interaction between the drug and the polymer used in this study. Thin layer chromatography and infra-red studies of the dispersed systems showed that the drug dispersions with Eudragit-RS® were stable. The in-vitro dissolution rate studies of the dispersed drug indicated a slower dissolution rate as compared with the pure drug.
