Abstract

The aim of writing this review on gastro retentive and floating drug delivery system was to compile the new literature with the principle mechanism of floatation to acquired gastric retention. Various attempts have been made to develop gastroretentive delivery systems such as high density system, swelling, floating system. In floating multiple unit and single unit system are design and their classification and formulation aspect is cover in detail. These systems are very helpful to different problem solve during the formulation of different dosage form. In this review the gastric physiology and reported intra gastric delivery system have briefly been presented.

Abstract

Hypertension is an important risk factor for cardiovascular disease, the incidence of which it is possible to reduce by prophylactic treatment with antihypertensive drugs. Adherence to long-term therapies for chronic diseases in developed countries averages only 50%. In developing countries the rate is even lower, probably reflecting inequalities of access to appropriate health care facilities, supervised by qualified health care professionals. In clinical practice, however, only a minority of patients reach target blood pressure levels. The resulting gap between actual and potential health gains has been attributed to the fact that many patients do not take prescribed treatment as recommended. Pharmacists can play important roles in helping their patients with high blood pressure manage their condition. Serving as a partner and coach, pharmacists can provide medication therapy management services, educate patients about Hypertension. Further Pharmacists, physicians and other healthcare professionals should regard patients as partners in all decisions on treatments involving medication.

Abstract

In this study, various matrix type transdermal patches containing carvedilol of variable combination of polyvinylpyrrolidone (PVP) and ethylcellulose (EC) were prepared by solvent evaporation technique. The patches were prepared by adding surfactant (sodium lauryl sulphate) to promote drug penetration which was found to increase transdermal transport. Therapeutic frequency ultrasound (1 MHz) and surfactant have been individually shown to enhance transdermal drug transport. In this study, we investigated the synergistic effect of ultrasound and surfactants on transdermal drug delivery. This study provides an investigation of the dependence of therapeutic-frequency sonophoresis on various ultrasound parameters, including ultrasound pretreatment time and the distance of the horn from the skin. Based on the in vitro release and in vitro permeation profile the formulation Fs1 (PVP/EC 1:2) shows the best release. Ultrasound was applied to pretreat the skin using a sonicator operating at a frequency of 1 MHz at an intensity 2 Watts/cm2. The optimum ultrasonication time of 50 min at a distance of 1 cm shows the best release. The skin conductivity enhancement was found to be inversely proportional to the distance of horn from the skin and it is directly proportional to ultrasonication pretreatment time. These findings will be useful in optimizing therapeutic-frequency sonophoresis.

Abstract

Adaptability is probably the most distinct characteristics of life which may be defined as sum of all non specific response of the body to any demands made upon it; fundamentally it was a physiological response; primary object of which was to maintain life & to re-establish the normal state. Immunomodulator activity of ethanolic and aqueous extracts of leaves and bark of Albizia lebbeck Benth. were investigated in Swiss albino mice by using swim endurance test and acetic acid induced writhing test model. The ethanolic and aqueous extracts of leaves and bark of Albizia lebbeck were administered to the experimental animals among which the ethanolic extract of Albizia lebbeck leaves have shown to be exhibit strong immunomodulator effect by increasing the swimming or survival time (P<0.001) and also decreased the writhing produced by glacial acetic acid (P<.001). The maximum increase in swimming or survival time was noted in mice receiving test and standard drugs which were significantly more than the control group animals. Test and standard drugs offered maximum protection against acetic acid induced writhes by reducing frequency of writhes per minute.

Abstract

The present study aims at investigating the anti-stress effects of cinnamon (Cassia zelynicum) bark extract in cold restrained stress (CRS) model. The cinnamon anti-stress effect was characterized using ulcer scoring, adrenal-gland weight, estimation of plasma glucose and cholesterol levels, as well as estimation of norepinephrine (NE) and corticosterone. Our results demonstrated that cinnamon is effective in minimizing stress responses by alleviating the increased level of ulcer scoring, plasma corticosterone, NE, glucose and total cholesterol levels which was induced in rats subjected to CRS. Thus, cinnamon is found to be effective in minimizing stress responses at a dose of 200 mg/kg thereby beneficial in stress therapy.

Abstract

Microwave-assisted organic synthesis is an enabling technology for accelerating drug discovery and development processes. Microwave organic synthesis opens up new opportunities to the synthetic chemist in the form of new reaction that are not possible by conventional heating and serve a flexible platform for chemical reaction. This review focuses on the advances in the developing of innovative application of microwave mediated synthesis. The efficiency of microwave flash-heating chemistry in dramatically reducing reaction times (reduced from days and hours to minutes and seconds) has recently been proven in several different fields of organic chemistry. The time saved by using focused microwaves is potentially important in traditional organic synthesis but could be of even greater importance in high-speed combinatorial and medicinal chemistry. The study presents examples that demonstrate the significance of these advantages to industrial application.

Abstract

Drug delivery in ocular therapeutics is a challenging problem and is a subject of interest to scientists working in the multidisciplinary areas pertaining to the eye. Most ocular diseases are treated by topical drug application in the form of solutions, suspensions and ointment. These conventional dosage forms suffer from the problems of poor ocular bioavailability, because of various anatomical and pathophysiological barriers prevailing in the eye. Various efforts in ocular drug delivery have been made to improve the bioavailability and to prolong the residence time of drugs applied topically onto the eye. The potential use of polymeric nanoparticles as drug carriers has led to the development of many different colloidal delivery vehicles. Indeed, the association of an active molecule to a nanocarrier allows the molecule to intimately interact with specific ocular structures, to overcome ocular barriers and to prolong its residence in the target tissue. This review discusses the physiochemical characterization, fabrication techniques, therapeutic significances, patented technology of Eudragit based nanoparticles and future possibility in the field of ocular drug delivery.

Abstract

In the recent years Microwave–induced Organic Reaction Enhancement (MORE) chemistry has now found an important utility values for carrying out chemical reactions in organic synthesis of compounds. This technique as an alternative to conventional energy sources for introduction of energy into reactions has become a very well-known and practical method in various fields of chemistry. Microwave-assisted organic synthesis is known for the spectacular accelerations produced in many reactions as a consequence of the heating rate, a phenomenon that cannot be easily reproduced by classical heating methods. Its specific heating method attracts extensive interest because of rapid volumetric heating, suppressed side reactions, energy saving, direct heating, decreased environmental pollutions, and safe operations Another area of interest which has been under focus recently is to avoid the use of organic solvent, which leads to wastage and is detrimental to the environment.

Abstract

Diabecon an ayurvedic formulation was tested for safety by estimation of heavy metal contents and also for acute and sub chronic toxicity. Heavy metal content in formulation were tested and found to be within limit as prescribed by WHO. In acute toxicity no toxic symptoms were seen at single oral dose of 2000 mg/kg, observed up to 14 days in rats. In sub chronic toxicity study, Diabecon was tested at the doses of 50, 250 and 500 mg/kg p.o. once daily for 90 days in rats. The results did not show evidence of any treatment related changes in body weight, food and water intake and ill health or behavioral changes, when compared with the control animals. Some significant changes were observed in high dose treatment group (500 mg/kg) in hematological and serum chemistry parameters compared to control group but were within the normal laboratory limit and were considered as incidental.

Abstract

In the present study, we have reported the synthesis of some novel heterocyclic derivatives comprising imidazole and 1,3,4-thiadiazole containing moiety. Imidazothiadiazoles are of interest because of their diverse biological activities and clinical applications. Reactions of biphenyl carboxylic acid with thiosemicarbazide in the presence of phosphorous oxychloride resulted in biphenyl containing 2-amino-1,3,4-thiadiazole which is then further subjected to condensation with α-bromoarylketone under reflux in dry ethanol. The structures of the newly synthesised compounds were characterized by various spectral techniques and screened for antibacterial activity against strains of Escherichia coli, Pseudomonas aeruginosa and Bacillus subtilis, and antifungal activity against Candida albicans, Saccharomyces cerevisiae and Aspergillus niger. The compounds exhibited moderate to good activity when compared with standards.

Abstract

Nine bacterial strains were isolated using xylan rich media. The bacterial strain KS09 was selected on the basis of qualitative and quantitative test. It was identified as Bacillus sp. via physiological, morphological and biochemical characterization. The xylanase was purified to homogeneity from crude extract of Bacillus sp. KS09 using ammonium sulphate fractioning and CM-Sephadex C-50. The final purification fold was 10.20 with a recovery of 36.18%. The enzyme was optimally active at 50°C, pH 7.0 and stable over a broad pH range of 6.0-11.0. The residual activity at 6.0-11.0 pH was 100% even upto 3 h of incubation. The enzyme showed 75, 70 and 60% thermal stability at 50, 55 and 60°C, respectively after 1 h of incubation. The kinetic parameters (Km 22.59 mM; Vmax 76.93 IU/mL) were estimated using Lineweaver-Burk plot for purified xylanase. The xylanase activity was inhibited by all the metal ions applied. The characteristic studies revealed that xylanase including its cellulase free nature, broad pH stability and temperature stability are particularly suited its industrial applications.

Abstract

Lacidipine (LCDP) is a dihydropyridine derivative categorized as an Anti-hypertensive Ca+2 channel blocker belonging to BCS class IV drug with low solubility and low permeability which presents a challenge to the formulation scientists. The development of a solid dispersion by solvent evaporation is a practically viable method to enhance dissolution of LCDP from oral dosage form. Solvent evaporation by Fluidized Bed Process (FBP) was the method of choice for SD as it improves wettability with simultaneous increase in porosity of granules resulting enhanced surface area producing higher dissolution rate and bioavailability of poorly water-soluble drug. Thus, the main object of the present invention is to provide stable pharmaceutical dosage form of LCDP with desired dissolution rate i.e. at least 80% drug release within 45 minutes, without use of disintegrant(s) and/or surfactant(s) or without micronization of the active ingredient per se. One more object of this invention is to provide a sophisticated robust process for the preparation of said pharmaceutical dosage form by Quality by Design (QbD) concept focusing on thorough understanding of the product and process by which it is developed and manufactured along with a knowledge of the risks involved in manufacturing by IRMA & FMEA study of the product with process and how best to mitigate those risks by developing design space with DoE & MVDA with outlined control strategy.

Abstract

The emerging field of metabolomics had profound importance in recent years as its wide applications in the field of drug discovery and drug development. With recent advances in the field of metabolomics it can be proved as a major tool in the process of drug discovery and development. In the agricultural/chemical industry, metabolomics may be used to develop herbicides and pesticides With increasing importance being placed on health and safety related aspects of our food, metabolomics can potentially be a valuable tool to monitor and improve the quality of what we eat; for example, in food processing and quality control, or in plant breeding for improved crop varieties and in the development of novel foodstuffs. The present review reveals the importance of metabolomics in drug discovery along with recent advances in the techniques of metabolomics along with its wide applications in various fields like drug discovery, drug toxicity profiling, food industries especially in food and allied beverages testing.

Abstract

Present article signifies the exigency for regulation and regulatory bodies involved in development of biosimilars. The principle for development of biosimilars included opting adequate reference product, manufacturing process optimisation, quality control procedure, preclinical and clinical studies. India’s first guidelines were enforced in 2012, with amendments in 2016. Thus, we elaborated the amended guidelines for development of biosimilars.
