Abstract

Objectives: Fine needle aspiration (FNA) continues to be an acceptable and reliable procedure for the preoperative diagnosis of breast lesions, particularly. The purpose of this study is to evaluate our performance with FNA cytopathology (FNAC) in specific subtypes of (lobular, papillary, medullary, mucinous, tubular and other) breast carcinomas. Methods: FNAC results of specified carcinomas of breast, cyto-diagnosed and subsequently biopsied in 2006-2011 were compared with final histopathological evaluation. The results of FNAC were interpreted as benign, atypical, suspicious, malignant and non-representative. The absolute and complete sensitivity rates, underestimation of malignancy rate and inadequacy rate of FNAC were correlated with histopathologic subtype. Results: Of cyto-histopathologically correlated 60 cases, 59 (98.3%) were females and 1(1.7%) was a male. Age range was 34-83 years with a mean value of 58.5(±13.3). Mean age was significantly lower (49) in medullary carcinoma patients. Of cases, 1(1.7%) was diagnosed as benign (C2), 5(8.3%) were atypical (C3), 10(16.7%) were suspicious (C4), 37(61.7%) were malignant (C5) and 7(11.7%) were inadequate (C1) by cytopathology. Of specified histopathologic subtypes, 36(60.0%) lobular, 8 papillary, 7 medullary, 5 mucinous, 3 tubular and 1 adenoid cystic carcinomas were included. Invasive lobular-type carcinomas had lower (52.8% absolute (C5)); 69.5% complete (C5+C4) sensitivity rates. The complete sensitivity rate was 100% in papillary, medullary, mucinous and tubular carcinomas. The underestimation of malignancy rate (13.9%) and false negativity (2.8%) was higher in lobular carcinomas while tubular carcinomas showed a higher inadequacy rate (33.3%). Conclusion: For FNAC of the breast in the preoperative diagnosis of specified breast carcinomas, lobular and tubular carcinoma subtypes limit cytodiagnostic yield of FNAC. As management of breast diseases necessitates triple approach (clinical, radiological, pathological), an awareness of the FNAC limitations by all specialists is important, especially when dealing with specified breast carcinomas to decrease false-negative and false-positive results.

Abstract

Objectives: The purpose of this study was to evaluate the serum lipid peroxidation in rats with experimental endometriosis who received etanercept (anti-tumor necrosis factor alpha) treatment. Methods: In this experimental study, 30 Wistar albino female rats were used. The surgical endometriosis model operation applied to 20 rats from those, in the 10's sham operation was performed in order to exclude the effects of anesthesia and surgery. Ectopic endometrial implants volumes of rats 4 weeks after the first operation were evaluated and blood samples were taken with a second operation. Then the rats were divided into 3 groups: Group I (did not receive any treatment, false operation, n = 10), group II (2 ml / day subcutaneous saline, endometriosis model, n = 10), group III (2 mg / kg / week etanercept subcutaneously 3 times, endometriosis model, n = 10). 3. After two weeks of treatment, ectopic endometrial implants volumes were evaluated with and second blood samples were taken from rats. Serum malondialdehyde (MDA) levels were analyzed by spectrophotometry. Results: The ectopic endometrial implant volumes of group III showed significantly lower results than group II in the post-treatment period (p =0.000). According to the same time group III implants showed significantly lower volume in the post-treatment period before the treatment (p = 0.005). In the post-treatment period, there was a significant difference between the groups in the MDA levels (p = 0.001), and group III was showed significantly lower levels of MDA compared to group II (p = 0.001). In addition, pre-treatment levels of MDA were also significantly lower compared to post-treatment period in group III (p = 0.028). Conclusion: In summary, these results suggest that etanercept has protective effects against endometriosis in rats, which may be attributed to attenuating oxidative stress.

Abstract

Aims: The aim of this study is to evaluate Mueller-Hinton Agar (MHA) as a simple medium for the determination of germ tube test (GTT). Methods: A total of 86 Candida species isolated from different clinical specimens were included. All isolates were cultured onto Sabouraud Dextrose Agar (Salubris, Turkey) and incubated at 37°C for 24-48 hours. They were identified by VITEK MS (Biomérieux, France). For serum GTT, a single Candida colony was inoculated into 0.5 ml freshly obtained human serum. For MHA GTT, an inoculum of single colony was streaked by Dalmau technique onto MHA and covered by a sterile coverslip. After incubation at 37°C for 1.5 h and 3 h, all sera and MHA plates were examined using a light microscope. Serum GTT was accepted as gold standard method for the determination of sensitivity and specificity values. Results: The identification results were as follows: 51 Candida albicans, one C. dubliniensis, 12 C. parapsilosis, 12 C. glabrata, 4 C. kefyr, 3 C. tropicalis, 3 C. krusei. Serum GTT was positive in 30 (58.8%) and 36 (70.6%) of C. albicans isolates at 1.5 h and 3 h, respectively. MHA GTT was positive in 21 (41.2%) and 40 (78.4%) of C. albicans isolates at 1.5 h and 3 h, respectively. All non-albicans species except C. dubliniensis were found as negative using both tests. Both GTTs were positive for C. dubliniensis at 1.5 h and 3 h. MHA sensitivity and specificity was determined as 58% and 92% at 1.5 h; 86% and 81% at 3 h, respectively. Conclusions: MHA is safer and easier medium for GTT that may be used as alternative to serum GTT. The evaluation of GTT on MHA after 3 h incubation is recommended since it has higher sensitivity compared to 1.5 h.

Today, these fundamental ethical requirements constitute the solid foundation for ethical principles. Because the principles of respect for individual autonomy, beneficence, do not harm/non maleficence and justice; obtaining informed consent for the autonomous decision of the participants, minimizing the harms and risks that may be encountered in the research, taking the necessary prevention for possible harms, create ethical duties towards fair treatment of the selection and preservation of participant's privacy.

Abstract

Schizophrenia is a unique disease which cannot be mimicked exactly in experimental animals. In experimental animals, the thoughts cannot be explained by talking like in men and this causes it difficult to form a sufficient schizophrenia model in experimental investigations. However, some models were used to investigate the effects of new drugs in the therapy of schizophrenia and in the physiopathology of schizophrenia. Some of these models can be classified as locomotion and stereotype models, conditioned avoidance responding, catalepsy test, rat paw test, selfstimulation paradigms, prepulse inhibition of the startle reflex, latent inhibition paradigms, evaluating schizophrenia by social interaction test in rodents, cognitive models, cellular and molecular models, pharmacological models, developmental models, genetic models.

3 Temmuz 1992 tarih ve 3837 sayılı yasa ile kurulmuş olan Kocaeli Üniversitesi’nde; 11 Temmuz 1992 tarih ve 21281 sayılı karar ile Tıp Fakültesi, 14 Ekim 1994 tarihinde 2547 sayılı yasayla ilişkili Lisansüstü Eğitim Öğretim Enstitülerinin Teşkilat ve İşleyiş Yönetmeliği uyarınca da Sağlık Bilimleri Enstitüsü kurulmuştur1,2. 16 Şubat 1995 tarihinde Derince Sopalı Çiftliği olarak anılan yerdeki Sosyal Sigortalar Kurumu Kocaeli Hastanesi’nin Kocaeli Üniversitesi ile ortak kullanımına ilişkin imzalanan protokol gereği Kocaeli Üniversitesi Eğitim, Araştırma ve Uygulama Hastanesi açılmıştır3. 1995-1996 eğitim-öğretim yılında Tıp Fakültesi’nde olduğu gibi Sağlık Bilimleri Enstitünde de eğitim- öğretime başlanmıştır. İlk kez yedi öğrenci Tıp Fakültesi’nin farklı anabilim dallarında, bir öğrenci de Beden Eğitimi ve Spor Yüksekokulu’nda olmak üzere toplam sekiz öğrenci lisansüstü eğitime kabul edilmiştir4. Eğitimde yapılan bu cesur hamleler bilimsel bir dergi çıkartılması yönünde de gösterilmiştir. Bu amaçla farklı bilim dallarına özgü birkaç dergi çıkartılmış, ancak hepsi uzun soluklu olamamıştır. Örneğin; 1995 yılında iki ayda bir çıkartılması planlanmış olan Tıp Fakültesi Aile Hekimliği Anabilim Dalı tarafından hazırlanan Aile Hekimi isimli dergi 3 sayı çıkartılabilmiştir5. Ocak 1997 tarihinde Kocaeli Üniversitesi Tıp Dergisi6 adıyla yayım hayatına başlamış olan dergi de ekonomik sebeplerle iki sayıdan fazla çıkartılamamıştır. Aynı yıllarda Fen Bilimleri Enstitüsü Dergisi çıkarma çalışmaları yapılsa da yayın hayatına geçiş olmamıştır7. Bunun aksine aynı yıl yayım hayatına başlamış olan Sosyal Bilimler Enstitüsü Dergisi, 1999 yılında Kocaeli Depremi nedeniyle bir süre yayımlanamamış olsa da 2000 yılından itibaren ulusal hakemli dergi olarak yoluna devam etmektedir. Yılda iki kez yayımlanan Sosyal Bilimler Enstitüsü Dergi’si elektronik ortamda da bilim dünyasına katkı sunmayı sürdürmektedir. Dergi ULAKBİM, ASOS, Akademik Dizin ve TEİ tarafından taranmaktadır8. 2008 yılında yılda iki kez olmak üzere Uygulamalı Yer Bilimleri Dergisi yayımlanmaya başlanmış ve başarıyla yayım hayatını sürdürerek, dergiye elektronik ortamda ulaşılabilirlik sağlanmıştır9. Mayıs 2013 yılında ilk sayısını yayımlamış olan Elektronik Mesleki Gelişim ve Araştırma Dergisi (Journal Of Occupational Improvement And Research- EJOIR), Hereke Ömer İsmet Uzunyol Meslek Yüksekokulu öğretim elemanları tarafından hazırlanan ulusal hakemli bir dergidir. Dergi, akademik bilgi ve becerilerin paylaşıldığı, disiplinler arası işbirliğini geliştirmeyi hedefleyen, doğa bilimleri, eğitim bilimleri ve sosyal bilimler alanında Türkçe ve İngilizce yazılan makaleleri kabul etmekte ve yılda iki kez elektronik ortamda yayımlanmaktadır10. 2015 yılı itibarıyla üniversitemizin iki dergiye daha kavuşması beklenmektedir. İlki, ulusal hakemli dergi olarak yayın hayatına başlayacak olan Görünüm adlı dergidir. Yılda en az bir defa elektronik ortamda yayımlanması planlanan dergide, Güzel Sanatlar alanındaki bilimsel yazıların yanı sıra, Sosyal ve Beşeri Bilimlerden, sanatla ilişkilendirebilen problemleri ele alan yazılara da yer verileceği bildirilmektedir11. İkincisi ise ulusal hakemli elektronik dergi olarak planlanan Sağlık Bilimleri Dergisidir. 2014 - 2015 Eğitim Öğretim yılı itibariyle 25 yüksek lisans, 13 doktora programı ile lisansüstü eğitimine önemli katkı sunmakta olan Sağlık Bilimleri Enstitüsünde, 2015/11 sayılı Yönetim Kurulu’nun 76. Sayılı kararıyla bir dergi çıkartılmasına karar verilmiştir. Yılda üç kez (Ocak, Mayıs, Eylül) elektronik ortamda yayınlanacak olan bu dergide öncelikle Üniversitemizde sağlık bilimlerinde yapılmış olan her türlü araştırmaya, derleme, olgu sunumu, mektup, bilim tarihi ve bilim felsefesiyle ilgili makalelere yer verilecektir12. Sağlık Bilimleri Dergisi’nin lisansüstü eğitim-öğretim yapmakta olan öğrencilerinin güvenilir bilimsel bilgiye ulaşılabilmesine katkı sağlamasının ötesinde, öğrencilerini bilimsel bilgiyi elde etmeye teşvik etme, bilgiyi hedef kitleye ulaştırmalarına ve paylaşmalarına yardımcı olma gibi temel amaçları da bulunmaktadır.

Abstract

Despite the many studies in the literature investigating the relationship between Internet Addiction (IA) and adolescent mental health, there are few studies on their problems with carrying out activities of daily living. The purpose of this descriptive study is to determine the IA status of adolescents (n=828) as well as some of the daily life activities that they postpone due to not being able to pry themselves away from the Internet. The IA scores of adolescents who sleep less than 6 hours, eat only two main meals, skip/postpone meals and have snacks at the computer instead, delay going to the toilet and taking care of hygienic needs (hand washing, bathing) while on the internet and have a poor academic standing because of postponing schoolwork as a result of being on the computer were higher (p<0.05). At the same time, it was found that these adolescents experienced pain in the back, neck, arm and other parts of the body, had eye problems (eye tearing, red eyes, pain) and digestive problems (constipation, gas) from sitting at the Internet for a long time. There were also significant differences between IA scores in terms of the adolescents’ medical complaints (pain, eye problems) and some of the emotional issues that they experienced (p<0.05).

Abstract

Objectives: TP53 gene is one of the most important tumour suppressor gene that mainly plays a role in DNA repair, cell apoptosis, cell senescence and cell cycle control. TP53 rs1042522 (Arg72Pro) polymorphism is nonsynonymous polymorphism that alters P53 protein capacity during tumour suppression. Based on the data above, the purpose of this study is to investigate association between TP53 rs1042522 polymorphism and breast cancer risk in hospital-based Turkish women population. Methods: After DNA isolation from peripheral blood of 508 breast cancer patients and 367 healthy women the genotyping for TP53 rs1042522 polymorphism in both groups was done by a PCR-RFLP method. Statistical analysis was done by the χ2 test with 95% confidence intervals and the Hardy-Weinberg equilibrium was confirmed for tested patient and control populations. Results: Genotype frequencies were 48.6%, 46.3% for GG, 40.7%, 44.7% for GC and 10.6%, 9.0% for CC in breast cancer and controls, respectively. There was not statistical difference between genotypes in cases and controls (χ2=1.591, P= 0.451). Allele frequencies for G allele were 69.0% in cases and 69.0% in controls, for C allele 31.0% in cases and 31.0 % in controls. Also, this results were found to be not statistically significant for G and C alleles (G allele: p=0.424, C allele: p=0.501). The distribution of TP53 rs1042522 genotypes was in agreement with HWE for controls (p>0.05). Conclusion: There is no statistically significant association between rs1042522 and the risk of breast cancer in studied hospital- based Turkish women population. For further, rs1042522 polymorphism may be associated with other genetic factors due to the interaction of p53 with various proteins that play role in programmed cell death. Also, the combined effect of TP53 polymorphisms and the genes such as cyclins, cyclin-dependent kinases and p21 which are related with cell cycle control can be studied or clinicopathological characteristics of breast cancer patients might be evaluated in terms of p53 rs1042522 genotypes in larger population size.

Abstract

Objective: The inflammatory responsiveness of the cells to Lipopolysaccharides (LPS) is commonly used for in vitro experiments. However, the correct dose of the LPS for cell line experiments is elusive. The LPS concentration that gives the maximal response is a critical point of in vitro inflammatory experiments. The aim of this study was to reevaluate the concentration dependent effect of LPS on the 293T human renal cell line. Methods: We evaluated the cell-detachment assay of LPS-stimulated 293T cell line monitored by xCELLigence Real-Time Cell Analyzer (RTCA) system. We applied increasing concentration of LPS followed by Roche xCELLigence Instrument based on the Real-Time Cell Analysis System. Results: Our results demonstrated that the 2, 4 and 8μg of LPS inhibit cell division which diverts cells to a steady-state phase, 1 μg acts as mitogen. Lower concentrations are no effect on cells. Conclusion: These work showed that LPS concentrations had various effects on cell proliferation and need to be estimated for each experiment before carry out the experiments.

Abstract

Objectives: In this study, which aimed to determine the healting behavior level of Physical Education and Sports teacher candidates. Methods: Therefore, the Health Life Style Orientation Scale was administered in 2013-2014 academic years. 84 students of the 4th year and 88 students of the 1st year; hence, total of 172 student studying in Physical Education and Sports Teaching Department of Kocaeli University. Results: In conclusion, healting life style behavior of the teacher candidates was not found significant in terms of the variables of age, gender, year of doing professional sports, participation in a leisure activity, year of university education, degree of parental education, parental residence of the last 5 years, place of accommodation as a student, whereas a significant difference was found in terms of the duration of doing amateur sports, and the economic status. Conclusion: This research has shown that healthy lifestyle behaviors are influenced by income level and participation in sports.

Abstract

Purpose: This study was conducted to explore the impact of the Mental Health and Psychiatric Diseases Nursing (MHPDN) course on the levels of approach-avoidance towards emotional situations of student nurses and on their problem-solving skills. Method: The study was conducted over the period September 2014 - February 2015 with fourth-year students in the Kırıkkale University Health Sciences Faculty and was of quasi-experimental design, using a posttest and a control group. The students taking the MHPDN course were recruited as the study group while the students in the other departments of the Nursing Faculty comprised the control group. A pretest and posttest was administered to the study group while the control group was administered only a posttest. The sample constituted 164 students who agreed to participate in the study. 44 students of the sample are study group and 120 students of the sample are control group. The researchers collected the data using a Personal Information Form, the Emotional Needs Scale (ENS) and the Problem-solving Inventory (PSI). The data collected were evaluated using descriptive statistical analysis, chi-square, ANOVA, Mann Whitney U and Wilcoxon tests. Results: Of the students, 43.9% reported that they had difficulty expressing their emotions and 56.1% said that they received emotional support from their family and relatives. The differences between the mean scores of the study group on the ENS pretest and posttest and on the sub-scale of "approach to emotions" were found to be statistically significant (p<0.05). It was also found that the differences between the mean scores of the study group students on the PSI pretest and posttest were statistically significant (p<0.001). The differences between the posttest mean scores of the study and control groups in the sub-scale "approach to emotions" were statistically significant (p<0.05). Conclusion: The results showed that the skills of the students taking the MHPDN course in terms of their approach to emotions had significantly increased while no statistically positive and significant difference was found in the perceptions of problem-solving. It is recommended that the problem-solving perceptions of students should be evaluated together with emotional needs and other factors, and training activities should be carried out to increase problem-solving skills.

Abstract

Cutaneous Acremonium infections in humans is rare. We report a patient with chronic lymphocytic leukemia who developed subcutaneous infection due to Acremonium sp. A wound lesion on distal wall of left lower leg was the first clinical manifestation of the patient. Fungal culture of deep tissue biopsy from the lesion revealed Acremonium species. In bacterial cultures Enterecoccus faecalis, E. clocea and Candida albicans were isolated. Pathologic investigation resulted as pyoderma gangrenosum and T cell lymphoproliferative lesion. Thus, the diagnosis of patient was considered as combined bacterial and fungal infection plus pyoderma gangrenosum. Thus, the patient was treated with ampicillin/sulbactam, methil prednisolone, topical naftifin and fucidic acid and itraconazole. The chemotherapy protocol was continued at the same time. Healing of the lesion was observed after ıtracanazoleteratment.

Abstract

Cancer is a fatal disease that interrupts human life unexpectedly. It is known as the most common reason for death following the deaths from cardiac disorders. In most people, the term “cancer” has been associated with death. Therefore, the psychosocial problems created by cancer diagnosis are inevitable. Normal response against cancer diagnosis varies from nervousness, anxiety, denial of the disease due to grief over the loss of health, and perceiving the progress of the disease as a battle or fatalistic acceptance. In the literatüre, the expression of a fatal disease has been termed as “breaking bad news”. Reporting the bad news to a patient is really a hard task for physicians because of the inability to predict how the patient and his/her relatives would react and lack of awareness on how to handle their reactions is a difficult situation for nurses and other health care personel. Generally, the experience and personality of the reporting nurse impress the informing approach. When we evaluate the explanation of the diagnosis, treatment, and progression of the disease to the patient, then we could determine whether our style of expression and our manner in order to meet the needs of the patient. Thus, this execute us the importance of education in communication skills. In this review, the ideal framework for reporting the process of telling the truth in cancer patients and nursing approach.

Abstract

Basing our diet on carbohydrates is determined by many research tasks. Carbonhydrate groups are present as structural and protective components in connective tissue of animal organisms and in addition they are involved in plant and bacteria cell wall as structural elements. They form glucoconjugates showing different characteristics on cell surface by covalent binding to lipids and proteins. These formations play a major role in such important events; intercellular recognition, cell-cell interactions and the interactions with the environment, cellular development and growth, cellular adhesion and regeneration. Carbohydrates spend 50% of the glycosylation of proteins in mammalian cells and take over some tasks of different proteins in the cell. Glycobiology the study of the vital importance of carbohydrate groups and their functions is investigating their role in the production of organisms. Glycobiology studies have determined the molecular structure of the sugar in glycoconjugates, the control of glycosylation patterns and the amount and copying carried out in the field of enzyme genes associated with phenotypic determinations. New strategies and techniques that study the structures and functions of carbohydrates are making strides toward creating more effective drugs with glycoproteins. Changes in the structure of glycoproteins and other glycoconjugates on the surface of cancer cells in cancer metastasis is thought to be important. Cancer cells in studies with the cell surface in the transformation of normal cells to cancer cells was found to have significant changes in the glycoconjugate. A tumour of the cellular defence system, by the use of certain substances capable of binding to this region, which undergoes changes in the surface of the cancer cells, may be more easily eliminated through quicker and better recognition. Protein-bound carbohydrates increase in tumour specimens of patients. Increases in the carbohydrate content of the normal protein, the glycoprotein of the tumour cells and production of new liver tissue or lymphoreticular tissues may be due to increased synthesis of glycoprotein. In this review, the structure of glycoconjugates and the glycobiologic approach in anti-cancer drug development and changes of cell surface components in cancer tissues were examined.
