Abstract
3,3’-Dimethoxybenzidine (DMOB) is a crystallinesubstance used to produce azo dyes and also asadherent agent and polyurethane elastomers ingredient.The dye and pigments DMOB-derivativeare used to paint leather, paper, plastic andrubber. Occupational exposure to this compoundoccurs during its synthesis and usage of azo dyes.In Poland, according to the data provided by theChief Sanitary Inspectorate, the number of employeesexposed to DMOB in 2005-2013 graduallyincreased and achieved 364 persons in 2013.There is a lack of data on acute poisoning of humansby DMOB. Symptoms of chronic intoxicationby this compound may be systemic hypoxiacaused by methemoglobinemia and hematuria.The oral LD50 value in rat indicates that DMOBis a harmful compound. In experimental animalsrepeatedly treated with this compound the increasein both liver and kidney weights, alterationsin thyroid gland, an increase in erythropoiesis inspleen, degeneration and also necrosis of hepatocytesin central zone of hepatic lobules were observed.DMOB induces point mutations in Salmonellatyphimurium, mutation in mice lymphoma cells,chromosome aberrations and SCEs in CHO cellsin vitro and chromosome aberrations in vivo.Carcinogenic effect of DMOB in humans was notobserved. In experimental animals the suffi cientevidence of carcinogenicity was obtained. IARCclassifi ed DMOB as a presumably carcinogenicagent for human (Group 2B).The recommended maximum exposure limit(MAC) for DMOB of 0.2 mg/m3 is based on therisk assessment of tumours appearance in malerats chronically exposed to this compound throughthe alimentary tract. On the basis of slopefactor (SF) value for the dose-response relationshipand relevant uncertainty factors the MACvalue at risk level of 10-4 was calculated. No STELand BEI values have been proposed. “Carc 2B”notation (carcinogenic substance) was proposed.
