A Novel SNP in the Coding Region of the Akirin2 Gene and Its Association with Marbling Score in Hanwoo (Korean Cattle)

Abstract
The Akirin2 is a nuclear factor which plays important roles in immune response, skeletal myogenesis, muscle development and carcinogenesis. Interestingly, a specific SNP located in 3'-UTR region of Akirin2 gene has previously been found to be significantly associated with marbling in beef cattle. Thus, the Akirin2 gene was considered as a positional functional candidate gene for marbling. The objectives of this study were to identify SNPs in the exonic regions of the Akirin2 gene and to evaluate the association of these SNPs with carcass and meat quality traits in Hanwoo (Korean cattle). In this study, I newly identified one exonic SNP (g.6559T>C) located within exon 1 in Hanwoo population. Genotyping of the SNP marker was carried out using PCR-RFLP analysis in Hanwoo steers to evaluate association with carcass traits. As a result, g.6559T>C SNP genotype was significantly associated with effects on marbling score. TT homozygous genotype or TC heterozygous genotype had a significantly higher marbling score (p<0.004) than those with CC homozygous genotype. Additive genetic effect was also observed for the marbling score (P<0.01) with higher marbling score of homozygous animals. Results from this study suggest that the exonic SNP g.6559T>C in the Akirin2 gene may be used as a DNA marker for the selection of Hanwoo with higher marbling.
