Analysis of the mineral nutrients content during the formation and maturation of somatic embryos of ‘FHIA-21’ (Musa AAAB)

Abstract

Despite the advances reached using somatic embryogenesis in Musa spp., there are not studies that state the requirements of mineral nutrient during its development. This investigation aimed to determine the mineral nutrient content in the culture medium at the beginning and end of the ‘FHIA-21’ somatic embryos formation and maturation phases. Two inoculation densities (0.5 and 1.5 gMF) of embryogenic cell aggregates were used for the somatic embryos formation in liquid culture medium. The number of somatic embryos formed was quantified after 30 days of culture. The content of Calcium, Magnesium, Potassium, Iron, Copper, Manganese and Zinc in the culture medium was determined by automatic absorption spectroscopy in both phases. Results showed the regulatory effect of inoculation density on the ontogenic formation and development of somatic embryos of ‘FHIA-21’. No relation between the inoculation density factor and mineral nutrients content was observed. The formation of a higher number of somatic embryos corresponded to a reduction in the content of Calcium, Magnesium, Potassium and Zinc. The importance of Iron in the maturation phase was evident.
