Apoptosis of primary osteoblasts induced by dexamethasone: cellular and molecular mechanisms
Journal Title: Eklem Hastaliklari ve Cerrahisi - Year 2005, Vol 16, Issue 1
Abstract
Objectives: We investigated the effect of glucocorticoids on cell viability of osteoblasts and explored the cytological and molecular mechanisms of osteoblast apoptosis induced by glucocorticoids. Materials and methods: Dexamethasone was used to induce apoptosis of primary cultured osteoblast-like cells from skulls of suckling mouse. Surviving rate of osteoblasts, apoptosis rate, activity of caspase-3, and DNA binding activity of nuclear factor-kappa B were assessed using MTT-dye reduction microassay, flow cytometry, colorimetric substrate assay and electrophoretic mobility shift assay. The surviving rates of osteoblasts and fibroblasts were also compared to assess the differences in dexamethasone-induced effects on cells. Results: Dexamethasone significantly decreased the survival rate of osteoblast-like cells through an apoptotic process, activated cellular caspase-3, and inhibited the activity of nuclear factor-kappa B, in a concentration- and time-dependent manner (p

Amaç: Bu prospektif çalışmada, lomber disk replasmanından (LDR) sonra komşu disklerde ve spinal segmentlerdeki faset eklemlerde meydana gelen değişiklikler araştırıldı. Hastalar ve yöntemler: Yirmi yedi olgu (7 erkek,...
Ranges of scapular elevation and depression in healthy male subjects
Objectives: Although most of the movements of the joints have been measured in several studies, data on scapular elevation and depression are very few. The aim of this study was to describe the amplitudes of active range...
Arthroscopic treatment of nontraumatic elbow ankylosis
Objectives: This study was designed to evaluate the results of arthroscopic treatment for nontraumatic elbow contractures with the removal of osteophytes and loose bodies of the olecranon process and olecranon fossa. Pa...
Spontaneous bilateral hip fractures following a seizure: a case report
Spontaneous bilateral hip fractures are very rare. They may occur secondary to several metabolic disorders. We report a 28-year-old male patient who developed spontaneous bilateral femoral neck fractures and a displaced...
The role of muscle integrity following midshaft clavicular resections: a cadaveric study
Objectives: There are conflicting reports in the literature regarding midshaft clavicular resections, and the role of muscle integrity has not been studied before. This cadaveric study was designed to investigate the eff...
