APPLE POLYPHENOL EXTRACT REDUCED JNK PHOSPHORYLATION IN THE LIVER OF INSULIN-RESISTANT RATS

Abstract

The apple fruits are the important source of biologically active compounds, such as flavanols (quercetin, kaempferol, and rutin), dihydrochalcones (phloretin and phloridzin), flavan-3-ols (epicatechin and procyanidins) and phenolic acids (caffeic acid and coumaric acid). Polyphenols extracts from fruits and peel of apples reveal high antioxidant, anti-inflammatory, antiproliferative activity. There are also a data on the antidiabetic activity of extracts. The aim of this study was to investigate the effect of apple fruits polyphenol extract (APE), as well as quercetin (Q) on blood serum glucose and insulin content, and liver c-Jun N-terminal kinase (JNK) activity under experimental insulin resistance (IR) in rats. IR was induced by keeping the animals on a high-calorie diet enriched with fructose (HCD) during 5 weeks. In animals from HCD group was observed the increase in blood serum glucose and insulin level. Also was fixed the phosphorylated JNK (p-JNK) content growth in liver tissue. The administration of APE and Q led to decrease in both glucose and insulin levels in blood as well as reduced p-JNK content in liver. The observed effects were more pronounced when APE was administered to animals. The obtained results indicate that APE revealed antidiabetic activity, normalizes glucose homeostasis and insulin signaling under experimental IR.
