بررسی رابطه پلی‌مورفیسم ژن PPP1R3 با دیابت نوع ۲

Abstract
زمینه و هدف: یکی از ژن‌هایی که اخیراً ارتباط آن با بیماری دیابت نوع ۲ تأیید شده، ژنPPP1R3 می‌باشد. این ژن روی بازوی بلند کروموزوم ۷ قرار دارد و رمزگذار پروتئین فسفاتاز 1 (PP1) بوده که دارای فعالیت سرین/ترئونین فسفاتاز است. در ناحیه 3'UTR این ژن، یک ناحیه پلی‌مورفیک وجود دارد که موجب ایجاد دو آلل ARE1 ,ARE2 میشود. این مطالعه با هدف بررسی ارتباط پلی‌مورفیسم ژن PPP1R3 با دیابت نوع ۲ انجام شد. روش بررسی: در این پژوهش مورد – شاهدی، ۱۰۰ بیمار دیابتی نوع ۲ و ۱۰۰ فرد سالم، به‌طور تصادفی از جمعیت مورد مطالعه انتخاب شدند. پلی‌مورفیسم ژن PPP1R3، با استفاده از روش PCR-RFLP مورد بررسی قرار گرفت. مقایسه متغیرها بین گروه سالم و بیمار با استفاده از آزمون تی‌تست، فراوانی آلل با شمارش و محاسبه نسبت آنها با آزمون کای‌دو و قرارگیری جامعه در تعادل هاردی – واینبرگ تأیید گردید. توزیع ژنوتیپ‌ها و آلل‌های مختلف در هر دو گروه سالم و بیمار با هم مقایسه شدند. یافته‌ها: در این مطالعه، تفاوت معنی‌داری بین فراوانی ژنوتیپ‌ها و فراوانی آلل‌ها در افراد مبتلا به دیابت نوع ۲ و افراد کنترل سالم مشاهده نشد. نتیجه‌گیری: نتایج این مطالعه نشان داد پلی‌مورفیسم در منطقه UTR'3 ژن PPP1R3 با دیابت نوع ۲، ارتباطی ندارد.
