Characterization of the Growth Hormone Gene (GH1) in Sudanese Kenana and Butana Cattle Breeds

Abstract
The aim of the present study was the identification of allelic variants of the bovine growth hormone 1 (GH1) gene in Sudanese Kenana and Butana cattle. A 2080 bp fragment spanning the region between 146 bp upstream of the first exon and 140 bp downstream of the last exon were sequenced in Kenana and Butana cattle. The sequence comparison of Kenana and Butana GH1 sequences with the reference GenBank sequence (M57764) descending from Holstein Friesian identified 10 and 12 single nucleotide polymorphisms (SNPs), respectively. Three novel SNPs were identified including g.1304C>T, g.2346C>T, and g.2537G>T (positions are given relative to the GenBank sequence M57764). A PCR-RFLP method was used to access the allele frequency distribution for the SNPs g.1059C>T (MspI restriction, silent substitution in exon 2), g.1547C>T (MspI restriction in intron 3), and g.2141C>G (AluI restriction, Leu/Val substitution in exon 5) in 61 and 54 animals of the Kenana and Butana breeds, respectively. Both breeds were found to be carrier of g.1059T allele with an allelic frequency of 0.35 for Kenana and 0.24 for Butana, whereas g.2141C allele was found to be fixed. The allele frequency of g.1547T (MspI (-) was highly significant (p<0.01) in Kenana (0.77) compared to Butana (0.59) cattle. The identified SNPs in Kenana and Butana cattle breeds could be used as genetic markers in different genetic studies.
