Comparison of Cell Culture, Integrated Cell Culture-PCR, and Direct PCR Methods for Detection of Infectious Bursal Disease Virus

Abstract
Infectious bursal disease represents the most severe poultry disease responsible for marked economic losses in Morocco. The diagnosis of this disease depends on the time and costly procedures. The aim of this first study in Morocco was to evaluate and compare the reliability of the methods of cell culture using two types of cells (Vero and BGM-70), integrated cell culture-PCR (ICC-PCR), and direct PCR for detection of infectious bursal disease viruses (IBDV) circulating in this country. A total of 55 bursal samples were collected from two different areas in Morocco and analyzed for viral contamination. In all the samples analyzed in this study, IBDV was detected in 37 (67.2%) of the samples, 53 (96.3%) samples and 31 (56.4%) samples who utilized cell-culture, integrated cell culture-PCR and direct PCR methods respectively. Our data demonstrate that there was a significant correlation (0.05 level) between these 3 methods to isolate IBDV. Further the sensitivity of ICC-RT-PCR method was evaluated, which indicated that this method is acceptable and sensitive enough to detect IBDV. Therefore, Because of this higher sensitivity and necessary efficiency needed to remove organic inhibitors, the ICC-RT-PCR method can be a suitable way for environmental surveillance and epidemiological assessment of infectious bursal disease in morocco.
