Computational prediction & Analysis of Fat mass and Obesity associated genes (FTO) responsible for Childhood obesity

Abstract
The childhood obesity is raising very critical problem among the children around the globe. More than 40 million children (<5 years) were overweight or obese in 2012 around the globe but it is preventable. Analysis of FTO gene had elucidated phylogenetic relations with other obesity related genes. While analysis of its pseudoknot structures (RNA) has revealed that it has different types loops within the structure and information about its conserved and unconcerned regions around the structure. The red regions of the structure suggest us that the same red region of the base pair can be matched and can be complementary to the other regions of the structure. SNPs predicted by self mode program at different regions that show mutations occurring in the FTO gene and the gene modifications taking place. FTO sequence revealed the ratio of the 132 nucleotides in the transversion mutation out of total 1627 nucleotides whereas the ratio of 141 nucleotides in the transition mutation out of total 1627 nucleotides. Among all the total nucleotides there were 5 silent mutations (SNPs) out of total mutation (278) occurred. Analysis of the FTO gene in s fold revealed the nucleotide regions of maximum probability, which could be targeted for in FTO gene causing drug resistance. Further analysis of the FTO gene with the help of a server STRING revealed about the protein-protein interaction taking place in FTO gene with other obesity related genes present around it. FTO gene and its protein interaction with other obesity genes was vital approach to get information about where protein-protein interaction is taking place. These conclusions about evolutionary relations of FTO gene with other related fat genes, its RNA pseudoknot structure, its SNPs, targeted regions of nucleotide positions and their protein-protein interaction deducted that we could target the specific regions of the nucleotide range. At these positions critical changes are being made which is the hot point of our targeted regions in the FTO sequence. The computational prediction server revealed the significance of FTO gene and once it could alter, the problem of obesity may get preventable in the children.
