Correlation between immune cell infiltration and patient outcomes in renal cell carcinoma

Abstract

Objective To use bioinformatics methods to analyze the relationship between different degrees of immune cell infiltration in tumors and prognosis of renal cell carcinoma (RCC), so as to provide a theoretical basis for clinical decision-making. Methods Gene expression profiles and clinical data of RCC patients were retrieved from The Cancer Genome Atlas database. CIBERSORT algorithm was used to quantify the tumor infiltration percentage of immune cells. According to the median of immune infiltration score as the cut-off value, the patients were divided into high infiltration group and low infiltration group of various immune cells. Kaplan-Meier curve was used to evaluate the relationship between immune cell infiltration in tumors and patient outcomes. Wilcoxon rank sum test was used to analyze the correlation between immune cell infiltration in tumors and clinical stage of patients. Results The infiltration of dendritic cells, mast cells, and activated CD4+ memory T cells in RCC was significantly increased (P < 0. 05). Compared with the low infiltration group, the survival of RCC patients with high infiltration of dendritic cells and mast cells was significantly prolonged (P < 0. 05), and the survival of RCC patients with high infiltration of activated CD4+ memory T cells was significantly shortened (P < 0. 05). In addition, with the malignant progression of clinical stage, the infiltration of dendritic cells and mast cells in RCC significantly decreased (P < 0. 05), and the infiltration of activated CD4+ memory T cells significantly increased (P < 0. 05). Conclusion The immune cell infiltration in tumors is associated with the prognosis of RCC. Increased infiltration of dendritic cells and mast cells suggests a better prognosis of RCC, and increased infiltration of activated CD4+ memory T cells suggests a poor prognosis.
