Crab Shell Extract Induces Prostate Cancer Cell Line (LNcap) Apoptosis and Decreases Nitric Oxide Secretion
Journal Title: Cell Journal(Yakhteh) - Year 2017, Vol 19, Issue 2
Abstract
Objective: Prostate cancer is the second most common cancer worldwide. Chemotherapeutic agents have been shown to have adverse side-effects, and natural compounds have been recommended for cancer treatment, nowadays. Crab shell has been shown to have cancer preventative and suppressive effects in vivo and in vitro. The aim of present study was to investigate the effect of crab shell extract on prostate cancer cell line (LNcap) in vitro. Materials and Methods: In this in vitro experimental study, LNcap cells were treated with different concentrations (0, 100, 200, 400, 800 and 1000 μg/ml) of crab shell hydroalcoholic extract in three different culture periods (24, 48 and 72 hours). LNcap viability was evaluated by trypan blue staining and MTT assay. Cell apoptosis and nitric oxide (NO) secretion were determined by TUNEL and Griess assays, respectively. Data were analyzed by one-way ANOVA test and P<0.05 was considered significant. Results: LNcap viability was decreased dose- and time-dependently. Thus 400, 800, and 1000 μg/ml doses showed significant differences compared to control group (P<0.001). Dose-dependent increase in the apoptotic index was also observed in 800 and 1000 μg/ ml concentrations (P<0.001). Nitric oxide secretion of LNcap cell was decreased time- and dose-dependently, while it was significant for 1000 μg/ml (P<0.05). Conclusion: Crab shell extract showed anti-prostate cancer effect, by inducing cell apoptosis and decreasing NO production.

Protective Effect of Aloe vera Extract against Bisphenol A Induced Testicular Toxicity in Wistar Rats
Objective: Bisphenol A (BPA), an endocrine-disrupting chemical, has been considered as a possible risk factor for fertility because it induces testicular toxicity. Thus, we sought to analyze the effect of Aloe vera as pl...
Urocortin Expression in Endometriosis: A Systematic Review
Objective Free radicals generated by ionizing radiation attack various cellular components such as lipids. The lung is a very radiosensitive organ and its damage is a doselimiting factor in radiotherapy treatments. Melat...
Evaluation of Mouse Oocyte In Vitro Maturation Developmental Competency in Dynamic Culture Systems by Design and Construction of A Lab on A Chip Device and Its Comparison with Conventional Culture System
Objective In conventional assisted reproductive technology (ART), oocytes are cultured in static microdrops within Petri dishes that contain vast amounts of media. However, the in vivo environment is dynamic. This study...
6-Formylindolo[3,2-B]Carbazole (FICZ) Enhances The Expression Of Tumor Suppressor MiRNAs, MiR-22, MiR-515-5p, And MiR-124-3p In MCF-7 Cells
Objective microRNAs (miRNAs) play bifunctional roles in the initiation and progression of cancer, and recent evidence has confirmed that unusual expression of miRNAs is required for the progress of breast cancer. The reg...
Orexin-A Modulates Firing Of Rat Rostral Ventromedial Medulla Neurons: An In Vitro Study
The rostral ventromedial medulla (RVM) acts a key role in the descending inhibitory pain modulation. Neuropeptide orexin-A (ORXA) is confined to thousands of neurons in the lateral hypothalamus (LH). While RVM gets the o...
