Detection of dsRNAs in Sclerotinia sclerotiorum isolates from Iran

Abstract

Sclerotinia sclerotiorum the causal agent of sclerotinia stem rot is one of the most important fungal plant pathogens with global distribution. Broad host range and causing severe damage on crops annually, has been increased the incentive for further study of this species. Nowadays possibility of using mycoviruses as modern strategies of fungal biological control is important. From agricultural perspectives, mycoviruses may contribute to sustainable agriculture as biological control agents. The presence of viral dsRNA were investigated in 50 isolates of S. sclerotiorum. Isolates obtained from different hosts including lettuce, sunflower, cabbage, rape-seed, soybean, tobacco and Artemisia in north of Iran. The dsRNA molecules were separated using STE buffer, Phenol, Chloroform and column chromatography of cellulose (CF-11). Out of 50, 23 isolates were detected which have dsRNA molecules. This is the first report of S. sclerotiorum dsRNAs from Iran. The impact of mycoviruses on the morphological characteristics of fungus was studied. The dsRNA-containing had less mycelium linear growth than dsRNA-free isolates.
