Detection of Peste Des Petits Ruminants Virus antigen by Reverse Passive Haemagglutination test: Application and Evaluation

Abstract

Reverse passive haemagglutination (rPHA) test was evaluated for detection of PPR antigen. rPHA test was compared with immunocapture enzyme linked immunosorbent assay (IcELISA) to determine the specificity and sensitivity. 94 ocular and nasal swab samples and 27 whole blood were collected from live animals suspected of PPR disease, while 111 pneumonic lung samples were collected from sheep and goats from recent PPR outbreaks and slaughter house in the White Nile state-Sudan. PPR antigen was detected in 8 (7%) lung tissue samples, 9 (33%) whole blood samples and 9 (9.6%) ocular and nasal swab samples by IcELISA, When using rPHA, PPRV antigen was detected in 17 (15%) lung tissue samples, 27 whole blood samples and 8 (8.5%) ocular and nasal swab samples. The agreement between rPHA and IcELISA for detection of PPRV was k=0.87 in lung sample, k=zero in whole blood samples and k=0.81 in swab samples. In conclusion reverse passive haemagGlutination was found to be cheap, easy to do, very sensitive and specific test to detect PPRV antigen in swabs and tissues.
