Dextran-Conjugated Anti-Immunoglobulin Antibodies: A Powerful Tool for Studying B Cell Receptor-Mediated Signaling
Journal Title: International Journal of Immunology and Immunotherapy - Year 2016, Vol 3, Issue 2
Abstract
Dextran-conjugated anti-immunoglobulin (Ig) antibodies (anti-Ig-dex) consist of a high molecular weight dextran (~1-2 × 106 MW) to which multiple monoclonal anti-IgM or anti-IgD antibodies are covalently linked. These soluble conjugates have been used in a process of discovery, in extensive studies of in vitro polyclonal murine and human B cell activation, proliferation, Ig secretion and class switching, through signaling via multivalent cross-linking of B cell receptors (BCR) on the B cell surface, in the presence of a multiplicity of added stimuli. The cross-linking of the BCR in a multivalent fashion represents a more physiologic means, relative to using unconjugated bivalent anti-Ig, for studying the functional consequences of B cell encounter with specific antigens in vivo. This reflects either the repeating epitope nature of polysaccharides, or the display of a given intact protein in multiple copies on the surface of follicular dendritic cells, macrophages and dendritic cells where they are encountered by B cells.
Visions of the Hereafter: Releasing the Brakes of the Immune System by Checkpoint Inhibition Immunotherapy
Activation and control of the immune system is regulated by costimulatory molecules as well as by checkpoint inhibitors. Checkpoints are essential in maintaining self-tolerance and minimizing collateral damage by modulat...
The Infectious Disease of the Immunocompromised Host and the Elderly: Listeriosis
Listeria monocytogenes is an uncommon cause of illness in the general population. However, this bacterium is an important cause of severe infections in neonates, pregnant women, the elderly, transplant recipients, and ot...
IL-1β, IL-6 and TNF-Alpha Secretion by PBMNC Stimulated with Polyclonal Antibodies Anti-Hrage (AGE-Like) is Enhanced by Hyperglycemia in Diabetes
Receptor for advanced glycation end products (RAGE) activation is known to play an important role in the development of diabetes complications by amplifying the inflammatory process. Herein, we examined the effect of pol...
Early Subcutaneous Immunoglobulin for X-Linked Agammaglobulinemia
Introduction: Subcutaneous immunoglobulin (SCIG) utilization is limited in Spain. Our aim was to describe our experience with SCIG from the diagnosis in a patient with X-linked agammaglobulinemia (XLA). Case Presentatio...
Fast Updosing of Subcutaneous House Dust Mite Immunotherapy is Well Tolerated by Pediatric Patients
Data on the safety of a cluster regimen to start a subcutaneous immunotherapy (SCIT) with house dust mite (HDM) extract in pediatric patients are scarce. This study investigates the rate of adverse events (AEs) and the a...
