Effect of Chemical Permeation Enhancers on the Transcutaneous Permeation of Alfuzosin HCl

Abstract
Objective: The present research is aimed to study the effect of chemical permeation enhancers on the transcutaneous permeation of alfuzosin hydrochloride. Methods: Alfuzosin containing HPMC gels were prepared using different chemical enhancers like dimethyl sulfoxide, sodium tauroglycocholate and ethanol at varying concentrations. The prepared gels were characterized for physicochemical parameters like drug content, rheological behavior and spreadability. The gels were subjected to ex-vivo permeation study using excised rat abdominal skin. FT-IR and histological studies were conducted for skin to have an insight on the permeation mechanism. Further the irritation potential of the formulations was also evaluated on rats. Results: The drug content was between 97 to 99% indicating uniform distribution of drug. The viscosity of the gel formulations was found to be between 3600-3900 cps. Among all the permeation enhancers tested, DMSO at a concentration of 10% (w/w) has shown an improvement in permeation when compared to control gel. The histological studies revealed a definite change in the structure of the stratum corneum with HPMC gel and also there was a clear difference in the FTIR spectra of untreated and alfuzosin gel treated stratum corneum suggesting the fluidization of the stratum corneum.Conclusion: Based on the investigations it is apparent that transdermal route offers an alternate route to deliver a therapeutically effective amount of alfuzosin hydrochloride.
