Effect of dentine bondings in combination with peroxide bleaching agents on the biosynthesis of DNA in human gingival fibroblasts

Abstract

Introduction. Tooth bleaching and filling with adhesive materials are common aesthetic treatment procedures during which oral soft tissues are exposed to the toxic effects of both peroxides and monomers from bonding systems and fillings. Complex interactions with oral mucosa of those reactive substances are not fully understood. Aim of the study. To evaluate the toxic effect of three dentine bondings in combination with carbamideperoxideCPandhydrogenperoxideHP on the biosynthesis of DNA in gingival fibroblasts. Material and methods. Heliobond, Adper Single Bond 2 and Xeno V, each in volumes of 5 μL and 10 μL, were tested after polymerization and at a dilution of 10-3. Then, 3% HP and 3.7% CP were added. The cytotoxicity was assessed on the DNA synthesis assay employing an inhibition of [3H]thymidine incorporation into DNA in the culture of HGF1 ATCC line. Results. Data showed weak antiproliferative effect of bonding agents and CP in fibroblasts culture. The profiles of DNA synthesis for bondings alone, excluding polymerized Heliobond in 10μL, were similar and positive. The combination of bondings with HP caused significant decrease in DNA biosynthesis to above 90%, while neither resins alone nor combined with CP exerted 50% reduction in DNA synthesis. Conclusion. The cytotoxic interaction of the dental material compounds may be the result of damaged DNA biosynthesis. Clinically, it seems preferable to use CP-containing products, which minimized this cytopathic effect. However, due to their potential cytotoxic activity no direct contact of these substances with the gums should occur.
