Effects of Different Rootstocks on Stoma Properties of Tubed (Covered) Saplings in 'Red Globe' Grape Variety

Abstract

This research was carried out in 2016 in order to determine the effects of different rootstocks (5BB, 110R, 41B, 1103P and 1613C) on stoma properties of tubed (covered) saplings in 'Red Globe' grape variety in 'Çalışkan Grapevine Nursery' found in Manisa province. For this purpose, five tubed (covered) seedlings were selected for each replication from each variety/rootstock combination, and then the stoma molds have been obtained from the slice tips of the leaves of 2nd, 4th, 6th and 8th nodes of the tubed seedling shoots according to the method of mold removing with the help of nail polish. The stoma molds were examined under a 10x40 magnified light microscope aim to determine stoma density and size, and then the numbers of stoma in an area of 1mm2 were counted by taking the 5.1 times (0.196 mm2 area of observation) number of the stoma. As the average of all nodes, the highest stoma density was obtained from the combination of Red Globe/1103P (177.20 number/mm2), the lowest stoma density from the combination of Red Globe/5BB (133.90 number/mm2), the broadest stomas from the combination of Red Globe/1103P (13.67 μm), the narrowest stomas from the combination of Red Globe/41B (12.54 μm), the longest stomas from the combination of Red Globe/1103P (25.74 μm) and the shortest stomas were obtained from the combination of Red Globe/41B (23.23 μm). Consequently, a regular decrease has been recorded in the stoma density and stoma size from 2nd to the 8th node of tubed seedlings.
