Effects of hyperglycemia and hypoglycemia on analgesic activity of Morphine in rat

Abstract

Administration of Glucose or Insulin alters the pharmacological effects of some drugs. It has been reported that alteration of blood Glucose level affects the animal sensitivity top Barbiturates and Amphetamine. In this study the effects of high and low blood Glucose concentrations on antinociceptive activity of Morphine were investigated. Male wistar rats weighing 200±25 g were used. Analgesic activity of morphine was assessed by tail-immersion method. Hyperglycemia was produced by injection of Glucose (1.25-5 g/kg, IP) or by induction of diabetes by Streptozocin (75 mg/kg, IP). Hyperglycemia was produced by one of the three methods: insulin Injection (0.2-1 IU/kg, IP), starvation for 24 hours or administration of oral hypoglycemia agents such as Chlorpropamide (125 mg/kg) or Glibenclamide (1 mg/kg). The results showed that glucose attenuated Morphine analgesia dose-dependently and diabetes had a similar effect. Hyperglycemia, regardless of its induction method, augmented the analgesic activity of Morphine. Since both Glucose administration and induced diabetes attenuated Morphine analgesia, it seems that this effect is not related to blood Insulin level. This is because, Glucose administration causes hyperinsulinemia, while diabetes resulted from destruction of β-cells of pancreas by Streptozocin causes hypoinsulinemia. Compared to Glibenclamide, Chlorpropamide had a more potent effect on augmenting Morphine analgesia. This may be due to Chlorpropamide stimulation of ADH release that is followed by an increase in B-endorphine levels
