Effects of quercetin on growth performance and intestinal antioxidant capacity and immune related gene expression in piglets

Abstract
[Objective] The experiment was conducted to explore the the role of quercetin in improving piglet stress syndrome by analyzing the effects of different contents of quercetin on piglet growth per-formance, intestinal antioxidant indexes, inflammatory cytokine secretion and tight junction protein gene expression. [Method] A total of 216 healthy DLY (Duroc × Landrace × Yorkshire) piglets with similar body weight (9.5±0.5) kg were randomly divided into 3 groups: the control group (basal diet), the Group 1 (basal diet supplemented with 200mg·kg-1 quercetin), and the Group 2 (basal diet supplemented with 400mg·kg-1 quercetin) for 15 days, with 6 replicates in each group and 12 piglets (6 male and 6 female) in each replicate. [Result] 1) Compared with the control group, the Group 1 and Group 2 could enhance the average daily gain (ADG) and reduce the diarrhea rate of piglets obviously; 2) Compared with the control group, the Group 1 and Group 2 could significantly improve the activity of glutathione peroxidase (GPx), superoxide dismutase (SOD) and catalase (CAT), and reduce the content of malondialdehyde (MDA) in the jejunum mucosa. Besides, the activities of GPx, SOD and CAT in the jejunum mucosa were increased significantly as the level of quercetin increased; 3) Compared with the control group, the Group 1 and Group 2 could greatly reduce the levels of pro-inflammatory factor of interleukin-2 (IL-2), IL-1β and tumor necrosis factor-α (TNF-α), and significantly increase the level of anti-inflammatory factor IL-10. Compared with other groups, the level of secretory immunoglobulin A (slgA) was significantly higher in the Group 2;4) Compared with the control group, the Group 1 and Group 2 could greatly increase the mRNA relative expressions of Claudin-1, Occludin, and ZO-2 in the jejunum mucosa. And the ZO-1 mRNA relative expression in the jejunum mucosa of the Group 2 was significantly higher than those of the other groups. Additionally, the mRNA relative expressions of Claudin-1 and Occludin in the jejunum mucosa of the Group 2 was greatly higher than that of the Group 1. [Conclusion] In summary, the quercetin supplementation could reduce the diarrhea rate of piglets, post weaning, improve the growth performance of piglets, enhance the intestinal antioxidant function, increase the secretion of anti-inflammatory factors and inhibit the secretion of proinflammatory factors, and promote the intestinal tight junction protein-related genes expression.
