Effects on Some Immunological Parameters and Gene Expression Levels of Lupin Meal (Lupinus albus) Replaced with Fish Meal in Rainbow Trout (Oncorhynchus mykiss) Diets

Abstract

In this study, the effects of different replacement levels of fish meal by lupin (Lupinus albus) meal (0%, ABU0; 15, ABU15;, 30, ABU30;, 45, ABU45 and 60 ABU60) were investigated in diet for rainbow trout (Oncorhynchus mykiss) in terms of TNF-α, IL-1ß and IL-8 genes expression levels and some immunological parameters. Test diets were formulated and prepared to contain 41.5% crude protein and 18.1% fat with fish meal and lupin meal as main protein sources. It was observed that the essential amino acid contents of the experimental feeds was higher than the requirement levels for trout. Blood and liver tissue samples from 15 fish from each group were taken at the end of the 60 days feeding experiment. Analysis of lysosome (LIZ), myeloperoxidase (MPO) and respiratory burst (RBT) activities were performed from the blood samples and βactin, TNF-α, IL-1ß and IL-8 genes, specific for rainbow trout expression levels, were measured from liver tissue samples. According to the results, expression levels of TNF-α, IL-1ß and IL-8 genes and LIZ and RBT activities were significantly increased (p<0.05), while MPO activities did not present any differences by dietary incorporation of lupin meal in rainbow trout feeds. As a result, it has been concluded that the use of lupin meal as a fish meal replacer in rainbow trout feeds might positively affect the immune system in rainbow trout fingerlins.
