FimH Typing to Investigate Sub Clone Variations in ST69, ST127 and ST131 of Uropathogenic E. coli (UPEC) Isolates

Abstract

Introduction: Multilocus sequence typing (MLST) used to characterize Uropathogenic Escherichia coli (UPEC) is not discriminatory enough because strains with identical sequence types (ST) may possess different genotypes which enable different life- styles. Therefore there is a need for a typing scheme with superior discriminatory power between closely related strains. The aim of this study is to provide a typing scheme for UPEC ST69, ST127 and ST131 which will provide superior discriminatory power among them. Materials and methods: Sequences of FimH genes detected in 41 UPEC isolates recovered from patients with Urinary tract infection at Central Manchester University hospital between 2007-2011 were used. Their MLST grouping already showed that 20 were of ST131, 10 were of ST127 and 11 were of ST69. Using their fimH sequences and reference sequence of E.coli fimH K-12, consensus sequences were formed. These consensus sequences were aligned to fumC and an alignment created. Pair wise comparison was carried out for phylogenetic tree construction. Molecular revolutionary relationships were examined by the neighbor-joining method. Results: The isolates were found to have originated from the same ancestral lineage. Every isolate in each ST was closely related to one another as indicated by the clusters they formed which consisted primarily of their respective STs. Conclusion: FimH and fumC sequence analysis used in typing UPEC isolates is useful in investigating and understanding the fine population structure of UPEC. It cannot represent standard MLST but when used in parallel with MLST, it gives higher discriminatory result than MLST alone.
