HEPATO-RENAL PROTECTIVE EFFECT OF ORGANIUM MAJORANA AGAINST PARACETAMOL TOXICITY IN RATS

Abstract
The present study investigated the hepato-renal effect of the Organium majorana leaves extract against the hepato-renal toxicity induced by paracetamol in female rats. Theirty five white female rats were used and divided into five groups each group contain 7 rats as a following: Group 1: Rats which served as the control was orally administered distilled water (the vehicle) for 30 consecutive days. Group 2: Rats in this group were received orally aqueous extract of organium (100 mg/kg), once daily for 30 consecutive days. Group 3: Rats in this group were received orally aqueous extract of organium (200 mg/kg), once daily for 30 consecutive days. Group 4: Rats in this group were served as paracetamol toxic control and were administered a single oral dose of paracetamol (2 g/kg), 1 h after last dose of distilled water administration. ats in this group were received orally aqueous extract of organium (200 mg/kg), once daily for 30 consecutive days followed by a single oral administration of paracetamol (2 g/kg), 1 h after the last organium dose administration. The degree of hepatoprotection was measured using liver enzymes (Aspartate aminotransferase (AST), Alanine aminotransferase (ALT), bilirubin, albumin, and lipid profile levels. The degree of renal protection was measured using creatinine, urea, total protein and glucose. Also, histopathological examinations of liver and kidney were done. The significantly disturbed liver and kidney functions by paracetamol toxicity were restored to nearly normal values by administration of Organium majorana extract. Also, the change in biochemical parameters in groups received orally paracetamol alone was modified towards the normal values in groups given paracetamol and Organium majorana extract. Histopathological effect of paracetamol on liver and kidney was also markedly decreased by co-administration of Organium majorana extract. Our findings concluded that Organium majorana extract possessed hepato-renal protective activity against paracetamol induced toxicity in rats and this protection may involve the reduction of oxidative stress.
