HPTLC METHOD OF QUANTITATION OF BIOACTIVE MARKER CONSTITUENT PINITOL IN EXTRACTS OF PISONIA GRANDIS (R.BR)

Abstract
Pisonia grandis R.Br. is an evergreen medicinal plant in the four O'clock family of Nyctaginaceae. Leaves, stems and root of this plant are extensively used by tribals in India to prepare several folk medicines to treat rheumatism, arthritis and diabetes. Recently pinitol has been reported from the leaves of Pisonia grandis and thus the present study was designed to quantify this antidiabetic molecule in the various extracts of leaves of Pisonia grandis by high performance thin layer chromatography (HPTLC). The calibration curve of a stabilized HPTLC method was linear in the range of 1.5 to 7.0µg per spot and the correlation coefficient was found to be 0.9718, thus exhibiting good linearity between concentration and area. The limit of quantification (LOQ) and limit of detection (LOD) was found to be 1.5 and 1.2 µg respectively. The present study also revealed that among all the extracts, PGSX3 and PGSX4 contain more than 8 µg of pinitol per milligram of extract and thus exhibited chloroform: methanol mixture (9:1) and 100 % ethanol are found to be effective solvents to extract pinitol from Pisonia grandis. This is the first report on quantitation of pinitol by HPTLC.
