Insight from "Calcium Paradox" due to Ca2+/cAMP Interaction: Novel Pharmacological Strategies for the Treatment of Depression
Journal Title: International Archives of Clinical Pharmacology - Year 2016, Vol 2, Issue 1
Abstract
Our discovery of "calcium paradox" phenomenon due to interaction of Ca2+/cAMP intracellular signalling pathways (Ca2+/cAMP interaction) may provide new insights for the treatment of psychiatric disorders, such as depression. This disorder is mainly resulting by reduction of serotonin and catecholamine release in central nervous system. Since 1975, several clinical studies have reported that administration of L-type Ca2+ channel blockers (CCBs) produces reduction in vascular resistance and arterial pressure in hypertensive patients, associated with an increase in plasma noradrenaline levels and tachycardia characterized by sympathetic hyperactivity. During almost four decades, these enigmatic phenomena remained unclear. In 2013, we discovered that this paradoxical sympathetic hyperactivity produced by CCBs is mediated by Ca2+/cAMP interaction. Then, the pharmacological manipulation of this interaction could be a more efficient therapeutic strategy for increasing serotoninergic and monoaminergic neurotransmission in depression.
Unacceptable Intravenous Administration of Chloral Hydrate Oral Solution
Chloral hydrate is very frequently used in medical procedures with the purpose of sedation. After oral or rectal administration, central nervous system (CNS) and respiratory depression, cardiac arrhythmia and gastric irr...
Increase in Carbamazepine Serum Levels in Patient Treated with Voriconazole
The addition of voriconazole to chronic treatment with carbamazepine 400 mg twice a day was associated with increased carbamazepine serum levels (14.1 mg/l) and serum concentration dose ratios. Three days after the voric...
Levothyroxine for Hypothyroidism: Current Status of Biowaiver and Bioequivalence Studies
Hypothyroidism is a common disease that needs to be closely treated, in order to reduce adverse symptoms and pathophysiological effects. For thyroid hormone replacement is preferred a therapy with a synthetic form of thy...
Dietary Flavonoids and Colorectal Cancer: Evidence from Epidemiological Studies
Identification of dietary agents related to colorectal carcinogenesis is of high importance to decrease the global burden of this severe disorder. As flavonoids abundantly present in plant-based food items are suggested...
Novel Concepts for Clinical Pharmacology from "Calcium Paradox" due to Neuronal Interaction between Signalling Pathways Mediated by Ca2+ and cAMP: from 1975 to 2017
The solution for the so-called "calcium paradox" has been revealed 4 years ago, when we demonstrated the involvement of the interaction between Ca2+ and cAMP signalling pathways (Ca2+/cAMP signalling interaction) in this...
