INVESTIGATION OF INACTIVATION OF CHITIN DEACETYLASE IN A MEMBRANE REACTOR

Abstract
Enzymes are protein that can lose its activity because of several physical and chemical factors. It was shown for several enzymes that mixing in a reactor, pumping could inactivate them. Using experimental set-up it is necessary to estimate the influence of these factors to choose conditions that will not inactivate an enzyme. In our experiments the influence of mixing rate and influence of elements of the experimental set-up on activity of chitin deacetylase were investigated. It was shown that the peristaltic pump (flow rate up to 300 mL/min) and the membrane module (transmembrane pressure up to 1.50 bar) did not inactivate chitin deacetylase, but the enzyme was sensitive on mixing in a batch reactor. Mixing rate above 300 r.p.m. decreased its activity
