Morphological characteristics of Pinus caribaea in vitro plants var. caribaea influenced by the use of sucrose in the multiplication phase

Abstract

The genus Pinus has been classified as recalcitrant in relation to in vitro root formation. This work was carried out to evaluate the influence of sucrose on the morphologic characteristics of Pinus caribaea var. caribaea in vitro plants. The effect of different IBA and inorganic nutrients concentrations was also evaluated in the rooting phase. The best results were obtained using 50 and 60 g l -1 of sucrose. A lower formation of new shoots and higher percentages of dry matter was achieved. More differentiated needles, very similar to those observed in natural conditions, were obtained adding 60 g l -1 of sucrose. Regardless the concentration of sucrose (30-60 g l -1 ) which allowed obtaining the in vitro plants, length of plants increased by increasing the IBA concentration in the rooting phase. For the percentage of dry matter, plants placed in 30 g l -1 of sucrose showed a decrement in dry matter when the IBA concentrations increased. The opposite was observed in plants placed in 60 g l -1 of sucrose. Plantlets coming from a subculture with 60 g l -1 of sucrose and transferred to a rooting culture medium with a 50% reduction of inorganic nutrients showed the highest percentage of cell wall residues (47.95%). These results demonstrate the importance to study the effect of sucrose and inorganic nutrient concentrations to obtain plants with a higher cell differentiation and ready to achieve in vitro root formation.
