First report of Achromobacter xylosoxidans on wheat rhizosphere in Iran and its biocontrol activity

Abstract
Achromobacter xylosoxidans belongs to betaproteobacteria, Burkholdriales and Alcaligenaceae. A. xylosoxidans NB1 isolated from wheat rhizosphere in Gorgan, Iran and some of its biocontrol aspects were assay. Biological control has potential for the management of many diseases. In order to Molecular identification, genomic DNA extraction was prepared with CTAB method, and a fragment of 16S rDNA of the bacterium was replicated with PCR that was carried out by using universal primers (27F and 1492R). After electrophoresis, PCR products sequencing was performed by Bioneer Company (South Korea). The nucleotide sequence was assembled with BioEdit, and compared with similar sequences available in NCBI and EzTaxon. Results showed that our strain was most similar to A. xylosoxidans zc1 (100%). Then this bacterium named and registered A. xylosoxidans NB1 with accession number KU504626 in gene bank. NB1 studied for mycelial inhibition of fungi and wheat seeds growth promoting. Biofilm assay showed that this strain has significant biofilm formation. A. xylosoxidans NB1, showed appreciable level of IAA and siderophore production ability. NB1 has good biocontrol activity on Curvularia lunata and Bipolaris sorikiniana and inhibit the mycelia growth and cause inhibition zone. This isolate didn’t have protease, lipase, HCN production and P solubility.
