Frequency and Spectrum of Mutations induced by Gamma radiations and Ethyl methane sulphonate in Sesame (Sesamum indicum L.)

Abstract

 The present investigation was undertaken to study the frequency and spectrum of chlorophyll and vital mutations induced by gamma radiations and EMS. About 5000 seeds of a local variety ‘LTK-4’ were got irradiated with 6 gamma radiation doses viz., 150Gy, 300Gy, 450Gy, 600Gy, 750Gy and 900Gy at BARC, Mumbai, to create variability. The seeds were also treated with three different doses of EMS viz., 0.5%, 1.0% and 1.5% EMS. The seeds were raised along with the control at CSKHPKV, Palampur and RSS, Akrot, Distt. Una (H.P.) during kharif, 2013. Each M1 plant was harvested individually and grown as plant to progeny row to raise M2 population at both locations during kharif, 2014. Observations were recorded on plant survival, chlorophyll mutants, plant height, number of primary branches per plant, number of secondary branches per plant, number of capsules per plant, capsules per axil, seeds per capsule and seed yield per plant. At Palampur, the chlorophyll mutation frequency on M2 family basis was highest in 150Gy dose followed by 450Gy and 600Gy. In chemical mutagen, the highest frequency was observed at 1.5% followed by 1.0% and 0.5% EMS. Five types of chlorophyll mutations could be identified and arranged in order of chlorina>viridis>xantha>albina> variegated. At Akrot, the chlorophyll mutants were maximum at 150Gy and decreased up to 450Gy dose consistently. In EMS, 1.0% obtained highest chlorophyll mutants followed by 1.5% and 0.5% EMS. The highest number of viable mutants were obtained in 150Gy dose followed by 300>450>600>750>900Gy at Palampur. In chemical mutagen, mutants were higher in 0.5% followed by 1.5% and 1.0% EMS. The gamma rays proved to be more effective in producing viable mutants as compared to EMS treatment. Both the mutagenic effectiveness and efficiency for chlorophyll and viable mutations decreased with increase in dose. Lower doses of physical and chemical mutagens have proved to be more effective and beneficial in inducing mutations in sesame.
