Novel mechanistic and clinical implications concerning the safety of statin discontinuation.
Journal Title: Pharmacological Reports - Year 2011, Vol 63, Issue 4
Abstract
The beneficial effects of statins have been discussed widely, and their preventative role has been confirmed in cardiovascular disorders, primary and secondary prevention settings, and in asymptomatic subjects with a high cardiovascular risk. Despite these benefits, discontinuation of statins is frequent in cardiac patients and might be associated with adverse outcomes in several conditions involving acute coronary syndromes or acute stroke. In this review, we focus on the mechanistic background of statins that might contribute to such negative changes and that extend beyond cholesterol-lowering effects, including the so-called pleiotropic statin activity. In particular, findings regarding the detrimental impact of statin withdrawal on endothelial function, inflammation, platelet activity or AT1 signaling are discussed, along with the possible clinical implications for statin safety.

Effect of selected antidepressant drugs on cytochrome P450 2B (CYP2B) in rat liver. An in vitro and in vivo study.
The aim of the present study was to investigate the influence of antidepressants with different chemical structures and mechanisms of action affecting serotonergic and/or noradrenergic systems - tricyclic antidepressant...
Effect of intraarticular tramadol administration in the rat model of knee joint inflammation.
Local administration of exogenous opioids may cause effective analgesia without adverse symptoms from the central nervous system. Experiments show that peripheral antinociceptive effect of opioids is observed especially...
Vinpocetine and piracetam exert antinociceptive effect in visceral pain model in mice.
The effect of vinpocetine or piracetam on thermal and visceral pain was studied in mice. In the hot plate test, vinpocetine (0.9 and 1.8 mg/kg), but not piracetam, produced a reduction in nociceptive response. Vinpocetin...
Repeated co-treatment with fluoxetine and amantadine induces brain-derived neurotrophic factor gene expression in rats.
In the present study, we investigated the influence of repeated treatment with fluoxetine (FLU, 5 or 10 mg/kg) and amantadine (AMA, 10 mg/kg), given separately or jointly (twice daily for 14 day), at the mRNA level (the...
