Plant regeneration of Dioscorea cayenensis subsp. rotundata Poir cv. ‘Blanco de Guinea’ from somatics embryos

Abstract

Despite the progress made in the production of yam (Dioscorea spp.) seed by both conventional and biotechnological methods, the low disponibility of planting material with physiological and sanitary quality continues to limit the production of this crop on a large scale. Somatic embryogenesis is an alternative for the propagation of plant material and an auxiliary tool for its genetic improvement. The present work was carried out with the aim of to regenerate D. cayenensis subsp. rotundata Poir cultivar ‘Blanco de Guinea’ lants from somatic embryos. The effect of three culture media on the maturation of somatic embryos and different combinations of 6-BAP and kinetin in MS culture medium for germinating somatic embryos was evaluated. The highest percentage of mature somatic embryos (36.66%) was obtained in MS culture medium with the salts at 50% of their concentration and without plant growth regulators, which favored their subsequent germination (60%) in MS culture medium with kinetin (1.0 mg l -1 ) and 6-BAP (2.0 mg l -1 ). The germinated somatic embryos were characterized by the presence of open leaves, well-defined stems with a dark green coloration and small white roots, which they continued their differentiation until forming complete plants at 60 days of culture.
