PLASMID PROFILING AND CURING OF METHICILLIN-RESISTANT STAPHYLOCOCCUS EPIDERMIDIS (MRSE) AND METHICILLINSENSITIVE STAPHYLOCOCCUS EPIDERMIDIS (MSSE) ISOLATES

Abstract

The minimum inhibitory concentration of antibiotics (penicillin and tetracycline) to two methicillin resistance S. epidermidis isolates had determined, they were 60U and 25μg/ml respectively for the isolate 32S, they were 40U of penicillin for the isolate 40S.Results from agarose gel electrophoresis showed two isolates of MSSE 5S and 51S don't have any plasmid while other two isolates of MRSE have plasmids, two plasmids found in S. epidermidis isolate (40S), one of them was large plasmid and the other was small plasmid and in S. epidermidis isolate (32S) contained three plasmids, one from these was large plasmid and the other two plasmids were small plasmids. The results of plasmid curing by using sodium dodecyl sulphate (SDS) and rifampin showed that the plasmids of the local isolates 32S and 40S was curred by treatment with 100μg/ml, 230μg/ml respectively from SDS and 150μg/ml, 200μg/ml respectively from rifampin.Cured cells from 40S isolate (MRSE) were sensitive to penicillin and still resistance to methicillin and the cured cells from 32S isolate were sensitive to penicillin, tetracycline and still resistance to methicillin.
