Role of SAP7 in Hypha Formation and Adhesion to Colorectal Carcinoma Caco-2 (ATCC)

Abstract

Secreted aspartic proteases (Saps) are considered as key virulence factors of [i]Candida albicans[/i]. Hopefully our outlook will widen the knowledge of [i]SAP7’[/i]s role in [i]C. albicans[/i] pathogenesis. The goal of our study was to investigate [i]SAP7[/i] expression during [i]C. albicans[/i] adhesion to intestinal human cells. Another objective was to study the role of [i]Sap8-10[/i] and transcriptional regulators: [i]EFG1[/i] and [i]CPH1,[/i] using the mutants: [i]Δsap[/i], [i]Δefg1[/i], [i]Δcph1 [/i]during growth on the Caco-2 monolayer. [i]SAP7[/i] expression was analyzed using RT-PCR; relative quantification was normalized against [i]ACT1[/i] in cells after growth on Caco-2. Adherence assay of [i]C. albicans[/i] to Caco-2 was performed in 24-well-plate. Results proved that [i]SAP7[/i] can play a role during the initial adaptation of [i]C. albicans[/i] to intestinal tract and it decreases over time. Up-regulation of [i]SAP7[/i] occured in the absence of [i]SAP8[/i] and [i]SAP10[/i] (genetic alternations dependence). [i]SAP7[/i] can be regulated by the morphogensis’ regulators during [i]C. albicans[/i] growth on epithelium. Adhesion of the mutants was indistinguishable from SC5314. The lack of neither Sap8-10 nor Efg1/Cph1 influences the adhesive behaviour of [i]C. albicans[/i]. Deletion of [i]SAP8-10[/i] resulted in no filamentation defects. The results help understand better the role of [i]SAP7[/i] during adhesion and morphogenesis in [i]C. albicans[/i].
