S0Synergistic effect of selected anticancer drugs with Vitamin – E against HT29 human adenocarcinoma cell lines

Abstract
The present study was investigate to study the synergistic effects of selected anti-cancer drugs i.e. 5-FU, IR and Vitamin-E (Vit-E) on induction of apoptosis in HT29 cells. GI50 values were determined by MTT assay and was found to be 25, 25μM for the individual compounds and it was found that at 200μM higher concentration of Vit-E has nosignificant cytotoxicity with HT29 cells. On combination of anticancer drugs with Vit-E has significantly reduced the drug dose with the higher cytotoxic efficacy. The light microscopic image revealed that on treatment with individual and combination of anticancer drug with Vit-E showed majority of HT29 cells had shrunken and rounded up from their culture surface when compared with control cells. Further the DNA ladder assay confirms the individual and combination of anticancer drug and Vit-E induced apoptotic genomic DNA fragmentation. In regard to kinetics of apoptotic cells induced by treatment, the maximum induction of apoptosis by the combined treatment with vit E was higher than that of individual anticancer drugs was proved using confocal microscopy bydual staining with AO/EB. Present study implicit the observation that Vit-E has promising anticancer ingredients in combination with IR and 5-FU. Moreover our findings demonstrate that incubation of anticancer drugs with Vit E resulted in significant cell growth inhibition, DNA fragmentation. Apparently, the promising active principles and underlying mechanism by which this activity was exhibited need to be further investigated.
