Safety of interferon β-1a for treatment of COVID-19ï¼ a real-world study based on FAERS database

Abstract
ObjectiveTo systematically evaluate the safety of interferon β-1a for treatment of corona virus disease 2019 ï¼COVID-19ï¼ï¼ and to provide references for interferon β-1a's clinical application.MethodsThis study was conducted with the database from US Food and Drug Administration adverse event reporting system ï¼FAERSï¼ from January 1ï¼ 2015 to March 31ï¼ 2021. Information component ï¼ICï¼ and reporting odds ratio ï¼RORï¼ methods were applied for signal mining.ResultsA total of 463 700 records of COVID-19 were selected for analysisï¼ and 45 positive drug adverse event signals were detected. Headache ï¼IC025=1.09ï¼ ROR025=2.28ï¼ï¼ pyrexia ï¼IC025=0.51ï¼ ROR025=1.51ï¼ and multiple sclerosis relapse ï¼IC025=3.67ï¼ ROR025=14.71ï¼ were positive adverse events with higher frequency. Autoimmune disorder ï¼IC025=4.42ï¼ ROR025=24.03ï¼ï¼ streptococcal infection ï¼IC025=4.12ï¼ ROR025=19.82ï¼ï¼ and multiple sclerosis relapse ï¼IC025=3.67ï¼ ROR025=14.71ï¼ were positive adverse events. Acute lung injuryï¼ cardio-respiratory arrest and metabolic acidosis were associated with a higher proportion and frequency of death.ConclusionThere are certain safety issues with interferon β-1a in the treatment of COVID-19ï¼ and some adverse events with high frequency and high death rate deserve further attention by medical staffs.
