STUDY OF THE IMMUNOHISTOCHEMICAL EXPRESSION OF p53 AND Ki67 (MIB1) IN OVARIAN TUMOURS AND THEIR CORRELATION WITH CLINICOPATHOLOGICAL FACTORS

Abstract

BACKGROUND Ovarian cancer, a leading cause of death, is characterized by marked heterogeneity in clinical presentation, so an accurate histopathological diagnosis is vital for management and to predict outcome of therapy. Approximately 50% of human malignancies present with mutations in p53, which is the most common tumour suppressor gene involved. Similarly, Ki67 protein is a cellular marker for proliferation. We wanted to determine role of p53 and Ki67 by IHC as a diagnostic and prognostic factor in the assessment of ovarian tumours and correlate with clinicopathological factors. METHODS Immunohistochemical staining was done for p53 and Ki67 by using Dako antibodies as per protocol. Positive staining in the nuclei of the cells, absolute number of cells stained, and intensity of the staining pattern were noted. RESULTS Out of the 110 ovarian tumours, surface epithelial tumour predominates (n= 76, 69.09%); followed by germ cell tumours (n= 24, 21.81%) and sex-cord stromal tumours (n= 6, 5.45%). We had 2 cases of monodermal teratoma (Struma ovarii) and 2 cases of secondary tumour (metastatic tumours). The p53% and Ki67 labelling index were seen to be high in malignant tumours of epithelial types (serous carcinoma) in advanced stages whereas it was low or absent in other tumours. CONCLUSIONS p53 and Ki67 helps in determining the grade of the ovarian tumours and are important prognostic factors in the assessment of ovarian tumours.
