The mechanism of electro-acupuncture at Shu and Mu acupoints of lung and large intestine meridians for relieving acute gastrointestinal injury in rats with acute pancreatitis

Abstract
Based on the theory of exterior-interior relationship between the lung and the large intestine, the mechanism of electro-acupuncture at Shu and Mu acuacupoints for relieving gastrointestinal injury of AP rats was explored. We found that electro-acupuncture at shu and mu acupoints of lung and large intestine meridians (BL13, BL25, LU1, ST25) can relieve AP rats gastrointestinal injury by rebalancing inflammatory and anti-inflammatory cytokines, and making higher level of DAO, VIP, CCK. Objectives: This study aims to explore the mechanism of alleviating gastrointestinal injury of rats with acute pancreatitis (AP) by electro-acupuncture at Shu and Mu acupoints of lung and large intestine meridians. Methods: All rats were randomly divided into 5 groups (n=40): group A [DaChengqi Decoction (DCQD), n=8], group B [Back Shu acupoints (BL13,BL25) and DCQD, n=8], group C [Front Mu acupoints (LU1,ST25) and DCQD, n=8], group D [Front Mu and Back Shu acupoints (BL13,BL25,LU1,ST25) and DCQD, n=8], and group E (the rat model of AP, n=8). The rat model of AP was established by retrograde infusion of 3% sodium taurocholate into biliary pancreatic duct. Excepting group E, all other groups were offered with DCQD by intragastric infusion within 24h after establishing models. In addition, group B, C and D received 3 times electro-acupuncture and 40 mins each time within 24 hours after establishing models, the heart blood, pancreatic tissue and intestinal tissue were took 12h after intragastric infusion to test the levels of tumor necrosis factor (TNF-α), interleukin 4 (IL-4), diamine oxidase (DAO), cholecystokinin (CCK), and vasoactive intestinal plypeptide (VIP). The pancreatic tissue and intestine tissue were taken for pathology observation.Results: Compared with group A, the level of TNF-α was lower and the level of IL-4 was higher in group B (P<0.05). Group B, C and D had higher CCK and DAO levels than group A (P<0.05). Group C had the highest CCK level of the three groups (P<0.05) and the level of DAO was better in group B, D (P<0.05). Group B, D had higher VIP level than group A and group B was the best of them (P<0.05).Conclusions: The mechanism of electro-acupuncture at Shu and Mu acuacupoints for relieving gastrointestinal injury of AP rats is associated with the rebalance between inflammatory and anti-inflammatary cytokines, and the increasing of DAO, VIP and CCK level.
