TRS-PCR based genotyping of Mycobacterium kansasii

Abstract

Introduction. In recent years increasing number of infections caused by NTM, including Mycobacterium kansasii, has been observed. For this reason it is important to find a reliable method, which would allow to lead epidemiological investigations.Material and methods. In our research 48 M. kasnasii strains were used, its PRA-hsp65 profiles were initially analyzed. Then we generated fingerprints using TRS-PCR method and analysed the repeatability of research results.Results and conclusions. 39 of 48 strains were classified according to PRA-hsp65 profiles as M. kansasii type I, which is the most commonly isolated type from humans. CGG-PCR and CCG-PCR tests proved to be the best method for our collection of M. kansasii, as they allowed for the greatest distinction between the strains. It was confirmed by high values of discriminatory indices, with repeatability at very similar levels. The TRS-PCR test has allowed us to confirm thePRA-hsp65 results for three strains, which according to PRA-hsp65 database were classified as M. fortuitum type I. These strains clustered separately from M. kansasii in all TRS-PCR analyses. Achieved results may suggest that TRS-PCR method has a potential to be used in epidemiological investigations of M. kansasii but only when is supported by other method.
