VIRUCIDAL AND ANTIVIRAL POTENTIAL OF L-AMINO ACID OXIDASE AND Β PROPIOLACTONE AND RELATED RIFT VALLEY FEVER VACCINE POTENTIALS

Abstract

Regarding the bioactivity of synthetic beta propiolactone (βPL) and natural L- amino acid oxidase (LAAO), derived enzyme from snake venoms, it was chance to study the effect as virucidal and antiviral agent for the expected re-virulence of chemical inactivated viruses. Recorded data revealed that βPL and LAAO has no antiviral activity while they proved ten virucidal activities against Rift Valley fever virus (RVFV).The results of RVFV post treatment with both LAAO and βPL showed a fast depletion rate 2.5 log (10) / hr while it was 1.25 log (10) / hr for βPL and LAAO respectively. Inactivants were used safely as no morbidity and mortality recoded 7 days post administration. Neutralizing antibodies was detected 15 days post vaccination and antibody (Ab) level was insignificantly different post vaccination with LAAO and βPL inactivated RVFV. Cellular immunity was traced and IFN-γ level was significantly elevated post immunization with LAAO than immunization with βPL inactivated vaccine and IL-5 was not significantly different post immunization with both βPL and LAAO inactivated RVFV vaccine candidates.
