Zastosowanie TLC i densytometrii do ilościowego oznaczania paracetamolu i kofeiny w złożonym preparacie farmaceutycznym

Abstract
Application of TLC and densitometry for the determination of paracetamol and caffeine in combined pharmaceutical formulation • The optimal chromatographic conditions allowing identification and also quantification of paracetamol and caffeine using TLC with densitometry in combined tablet dosage form has been developed in this work. The studied sample was ethanolic extract coming from tablets consisted of 500 mg paracetamol and 65 mg of caffeine per tablet. Off all chromatographic conditions tested, the mixture composed of chloroform-acetoneammonia (25%) in volume composition 39.6:9.9:0.5 and silica gel 60F254 plates with concentrating zone were the most effective in the separation of both examined substances and compounds related to paracetamol. Densitometric detection was carried out at wavelength λ=250 nm for paracetamol and λ=275 nm for caffeine. Validation of proposed TLC method combined with densitometry confirms its specificity, precision, accuracy, robust and sensitivity. The results obtained in this work show that the developed method can be applied in routine control of preparations containing both paracetamol and caffeine.
