Aspects of pharmacokinetics of Cidipol drug while rectal and dermal application as basis for its clinical use

Abstract
Cidipol is a combination drug in the form of a solution of chlorinebrown aldehyde in dimexide and polyethylene glycol (PEG-400). Objective — to study the pharmacokinetics of Сidipol drug while rectal and dermal application, as also to evaluate the features of its metabolism in the body of animals. Materials and methods. Experiments were carried out on rabbits of the chinchilla breed (six animals for each route of administration) in which blood plasma was examined by gas chromatography (GC) at certain intervals after the administration of the drug. Results and discussion. The pharmacokinetic profile of the drug during external application to animals was characterized by the presence of a half-hour lag (hidden) period. Then slow absorption of ciminal with a maximum accumulation (Tmax) of 3 hours, its slow elimination from the body of an animal with a half-life (T0.5) of 5.3 hours and elimination coefficient (Kel) 0.13 were recorded. The volume of distribution of ciminal (Vp) with external application of Cidipol is 10 500 ml. When administered rectally, its pharmacokinetic profile is characterized by a slow absorption of ciminal with a semiabsorption period of 1.1 hours, Tmax = 2.5 hours and a rather slow elimination from animals’ body (T0.5 = 5.1 hours, Kel = 0.13). Vp of a ciminal makes up 3600 ml, which testifies to its ability to accumulate in tissues in higher concentration than in blood. Conclusions. It is established that the time of transmembrane diffusion of PEG-400 and the time of semisuscitation of the ciminal are almost similar, that is, overcoming the membrane barriers of ciminal occurs in the local environment of its solvents, and the preparation Cidipol with rectal administration and application of the skin is absorbed with solvents as a whole. Accumulated Vр of the drug in the skin and on the mucous membrane of the rectum are evaluated as positive for the implementation of the pharmacological effect. The results obtained provide a good reason for the clinical use of Cidipol in dermatological and cosmetological practice, as well as in pathological processes in the rectum.
