Attenuation of Allodynia and Hyperalgesia by the Leaf Extracts of Clerodendrum Formicarum in Rat Model of Diabetic Neuropathic Pain

Abstract

Painful neuropathy is the most common and debilitating complication of diabetes and results in allodynia and hyperalgesia. Hyperglycemia clearly plays a key role in the development and progression of diabetic neuropathy. Current therapeutic approaches are only partially successful and they are only thought to reduce the pain associated with peripheral neuropathy. The effective therapeutic management of this disease is pharmacological. However, the use of these drugs is limited by numerous adverse and side effects. Some natural products offer antinociceptive properties that may help to treat in a more integrative manner this disease. The objective of this study was to evaluate the effects of Clerodendrum formicarum on allodynia and hyperalgesia. Rats were injected with 150 mg/kg Alloxan monohydrate, to produce hyperglycemia. The Von Frey filaments test and the pressure test on paw were used to assess activity of Clerodendrum formicarum extracts. After induction of diabetes, mechanical allodynia and mechanical hyperalgesia was observed in rats. Administration by subcutaneous (sc) leaves extracts Clerodendrum formicarum (100, 150 and 200 mg / kg) attenuated the mechanical allodynia and mechanical hyperalgesiadose-dependent manner in diabetic rats. The anti-allodynic and anti-hyperalgesic of Clerodendrum formicarum was similar to that produced by pregabalin (10mg / kg, sc). These results suggest that an alternative is possible in the therapeutic management of painful diabetic neuropathy.
