CHEMO-SUPPRESSIVE ACTIVITIES OF CLOTRIMAZOLE AND GABAPENTIN IN A MURINE MALARIAL MODEL

Abstract
AIM: To assess the chemo-suppressive activities of clotrimazole and gabapentin in a murine malarial model by four-day suppressive test. STUDY DURATION: This study was done between June and December, 2017. METHOD: Clotrimazole (CLT), gabapentin (GBP), combination of clotrimazole and gabapentin (CLT+GBP), dihydroartemisinin-piperaquine (DHP) and DMSO 5% v/v were administered to infected mice, from 24 hours post parasitic inoculation respectively. Average percentage parasitemia (%parasitemia) and percentage chemo-suppression (%chemo-suppression) were calculated separately for day-4 and day-7. RESULTS: Each of treated groups had longer survival days and significant (p<0.001) reduction in average parasitemia on both days when compared to negative control group. CLT+GBP had significant (p<0.001) parasitemic reduction on both days when compared to either of CLT or GBP. CLT had significant (p<0.001) lower parasitemia on both days when compared to GBP. DHP showed significant (p<0.001) lower parasitemia on day-4 when compared to CLT+GBP but both had no parasitemia on day-7. Percentage chemo-suppression on day-4 versus day-7 was 76 versus 99%, 75 versus 99%, 96 versus 100% and 100 versus 100% for CLT, GBP, CLT+GBP and DHP respectively. CONCLUSION: CLT+GBP showed higher chemo-suppressive activity than either of CLT or GBP on both days and equal antimalarial potential to a standard fixed combination drug, DHP on day-7.
