Chromosome Karyotype Analysis of Transgenic Chickens

Abstract

The effects of exogenous gene p53 on the chromosome of transgenic chickens were investigated. The transgenic roosters with exogenous gene p53 were used as the experimental group,and the same breed with non-transgenic roosters were used as the control group. Chromosome specimen were prepared by peripheral blood lymphocytes culture method and bone marrow method. Fifty cells with clearly dispersed chromosomes were randomly selected to measure the chromosomes number,relative length,arm ratio and centromere index via oil immersion. The chromosome morphology between the transgenic and non-transgenic chickens were compared. The results show that the chromosome numbers of cocks in the experimental group and control group are in line with 2n = 78 which both contain 10 pairs of large chromosomes and 29 pairs of tiny chromosomes,and the tiny chromosomes are basically telocentric chromosomes. The chromosomes karyotypes in the two groups contain the 38 pairs of autosomes and one pair of sex chromosome ZZ. Chromosome 1,2 and the sex chromosome ZZ are the central centromere( m) chromosome. Chromosome 3 is the telocentric( t)chromosome. Chromosome 4 is the sub-central centromere( sm) chromosome. The chromosomes from No. 6 to No. 10 are telocentric( t) chromosomes.
