Cleared-ovule method applied to the evaluation of the effectiveness of pollinations in wide crosses within Brassica genera

Abstract

An attempt was made to adapt a cleared-ovule method developed by Ponitka and Ślusarkiewicz-Jarzinę (2004) for evaluation of crossing effectiveness in wide crosses within Brassica genera. The material used in this experiment consisted of 10 and 15 day old siliqua obtained after pollination of winter and spring oilseed rape (B. napus), Ethiopian mustard (B. carinata) and yellow seeded spring turnip rape (B. campestris ssp. trilocularis, cv. Yellow Sarson) with pollen of different Brassica species treated by different doses of UV radiation. Obtained data showed that cleared-ovule method elaborated previously for cereal crops can be successfully applied for the observation of Brassica embryo development after some small modification, instead of applying a time-consuming paraffin method. Effectiveness of interspecific crosses was considerably lower as compared to intraspecific crosses. Moreover in reciprocal interspecific crosses different effectiveness was observed depending on the maternal form.
