Cloning and Subcellular Localization of ARGONAUTE1 Gene from Populus euphratica

Abstract
The full length open reading frame (ORF) of homologous of AGO1 (AtAGO1) in Arabidopsis from Populus euphratica was cloned by using reverse transcription-polymerase chain reaction (RT-PCR) method based on Populus genome database.Sequencing results show that the ORF of the gene is 3 180 bp, and the ORF encodes a protein which is composed of 1 059 amino acids residue.The protein has 79.51% similarity compared with AtAGO1 amino acid, and the protein is named as Pe AGO1.The theoretical isoelectric point and molecular mass of PeAGO1 are 9.45 and 117 ku, respectively.Pe AGO1 does not contain signal peptide sequence, and it is speculated that PeAGO1 is not an exocrine protein.Phylogenetic analysis suggests PeAGO1 protein of Populus euphratica is relatively close to woody plants such as Prunus persica, Vitis vinifera L.and Eucalyptus grandis Hill ex Maiden in evolution, and not close to monocotyledon such as Zea mays L.and Oryza sativa.The cell location detection vector 35 S::GFP-PeAGO1 was constructed, and was instantaneously expressed in leaves of 84 K Populus by using protoplast conversion method.The results of laser scanning confocal microscope show that Pe AGO1 protein is localized in the nucleus.
