Comparison of molecular markers ( RAPD, ISSR) for determination genetic differences two of Crocus genus

Abstract

In this study, the genetic diversity of 25 Iranian Crocus genotypes from three species (C. speciosus, C. cancellatus and C. sativus) were evaluated using ISSR and RAPD molecular markers. 19 RAPD primers were selected to produce polymorphic and high resolution bands. In total, 261 DNA fragments were polymorphic and 13 ISSR primers were selected in this study. In total, 161 DNA fragments were obtained, from which 153 were polymorphic. Based on the Mantel test, significant correlation was found between similarity matrixes of RAPD and ISSR markers (r = 0.34).
