Determination of acute toxicity parameters and improving the effect of utero-tonic preparation “Uterodev”

Abstract

Introduction. The designed new injectable preparation “Uterodev” recommended for the stimulation of labor activity, atony and hypotension of the uterus, prophylaxis of postpartum complications (postponement delay, MMA syndrome, etc.), complex treatment of endometritis and metritis, fertilization improvement, obtaining embryos from donor cows, sows, sheep and goats should undergo preliminary tests on laboratory animals according toxicological parameters. The goal of the work was to determine the parameters of acute toxicity and improving the effect of utero-tonic preparation “Uterodev” on laboratory animals. Materials and methods. The study of acute toxicity of the injectable preparation “Uterodev” with the proparanolol hydrochloride active substance was performed on 50 white inbred rat’s males and females weighing 190–220 g, kept in vivarium conditions. They were divided in experimental and control groups. The preparation was introduced intragastric in the dose ranged from 5000 to 25000 mg/kg of live weight. Calculation of median lethal dose (LD50) and other indicators was determined by Litchfield and Wilcoxon method modified by Roth (1963) and the method described in the publication "Preclinical studies of veterinary medications" (2006). Results of research and discussion. Signs of intoxication in rats were not observed. Reflex excitability in all animals has been preserved. During experiment, deaths of the animals were not detected within 2 weeks. According to the research on “Uterodev” acute toxicity parameters, the LD50 at intragastric administration into rats accounted for above 5000 mg/kg of body weight. Thus, the preparation “Uterodev” corresponds to the fourth class of danger (according to GOST (National State Standard) 12.1.007-76). At “Uterodev” acute irritation, tests of the on the skin of guinea pigs there were not recorded erythema, scapular or edema in the animals. For the entire period of observations, thus the index of direct skin irritation was 0 points for the investigated object. Conclusions and prospects for further research. According to the research of “Uterodev” acute toxicity parameters it was set the value of LD50 at intragastric administration to rats was above 5000 mg/kg. This allowed attribute this preparation to the fourth class of danger. Investigations of acute irritant activity shown that “Uterodev” did not cause direct skin irritation in animals. Further studies will to be focused on the definition of chronic toxicity, embryotoxic and teratogenic properties of “Uterodev”.
